摘要
探索青海血蜱雌、雄成蜱唾液腺中蛋白的调控和变化规律,用非标记定量蛋白组学分析,联合液相色谱-质谱(LC-MS/MS)技术,鉴定分析青海血蜱雌、雄成蜱唾液腺组织中的蛋白质,筛选出差异表达蛋白,对差异表达蛋白进行基因本体论(GO)、京都基因和基因组百科全书(KEGG)和结构域(IPR)相关功能富集分析,亚细胞定位以及蛋白互作网络(PPI)分析。结果显示,在青海血蜱雌、雄成蜱唾液腺组织中有506个上调蛋白和447个下调蛋白。GO富集分析表明,差异表达蛋白主要富集在对刺激的反应,KEGG通路主要富集在嘌呤代谢,结构域主要涉及神经递质门控离子通道等结构域,差异表达蛋白主要定位于细胞核。PPI分析发现关联度最高的蛋白被注释为细胞色素P450(CYP450)。揭示了青海血蜱雌、雄成蜱唾液腺蛋白表达的差异,有助于更深入了解蜱的生理功能和调控机制,为抗蜱疫苗的研发提供新思路。
To explore the regulation and change of proteins in salivary glands of female and male adults of Haemaphysalis qinghaiensis,proteins in salivary glands of female and male adults of Haemaphysalis qinghaiensis were identified and analyzed using Label-free proteomic analysis combined with liquid chromatography-mass spectrometry(LC-MS/MS).Differentially expressed proteins were screened and subsequently subjected gene.The results showed that there were 506 up-regulated proteins and 447 down-regulated proteins in the salivary glands of female and male adults of Haemaphysalis qinghaiensis.GO enrichment analysis showed that the differentially expressed proteins were mainly concentrated in response to stimulus,the KEGG pathway was mainly concentrated in purine metabolism,and the domains mainly involved in the neurotransmitter-gated ion channel,and the differentially expressed proteins were mainly located in the nucleus.PPI analysis found that the protein with the highest correlation was annotated as cytochrome P450(CYP450).This study revealed the differences in salivary glands protein expressions between female and male adults of Haemaphysalis qinghaiensis and provided new ideas for the development of anti-tick vaccine.
