不同橘皮提取物对S.aureus和E.coli的抑菌活性对比研究及其抗菌机制初探

Comparative study on the antibacterial activity of different orange peel extract against Staphylococcus aureus and Escherichia coli and preliminary exploration of their antibacterial mechanisms

摘要 以橘皮为研究对象,探究了不同提取方法制备得到的橘皮提取物对S.aureus、E.coli抑菌活性的影响。采用二倍稀释法进行活性测定,酶标仪测定橘皮提取物对S.aureus的生长曲线,通过检测碱性磷酸酶的含量变化、激光共聚焦实验,评价橘皮提取物对S.aureus细胞壁、细胞膜的影响。结果表明,提取条件为水提,60℃,提取2次,3 h+2 h,料液比1∶5,pH为5~6时酶解,再酸解后乙酸乙酯和正丁醇萃取制备的橘皮提取物对S.aureus、E.coli的抑制效果较好。抗S.aureus作用机制表明,橘皮提取物能够改变S.aureus的细胞膜及细胞壁通透性,从而杀伤S.aureus。综上,不同的制备方法对橘皮提取物的抑菌活性有显著影响,为探究橘皮提取物抑菌活性提供了参考。 Using orange peel as the research object,the effects of different extraction methods on the antibacterial activity of S.aureus and E.coli were investigated.The activity was determined by double dilution method.Using an enzyme standardization to determine the effect of orange peel extract on the growth curve of S.aureus,and the effects of orange peel extract on S.aureus cell wall and cell membrane were evaluated by detecting the change of alkaline phosphatase content and laser confocal experiment.The results indicate that the extraction conditions were:After water extraction,60℃,2 times extraction,3 h and 2 h,solid-liquid ratio 1∶5,pH 5~6,enzymatic hydrolysis,and then acidolysis,the orange peel extract prepared with ethyl acetate and n-butanol extraction has the better inhibition effect on S.aureus and E.coli.The results showed that the extract of orange peel can change the permeability of cell membrane and cell wall of S.aureus,and thus kill S.aureus.In summary,different preparation methods have significant effects on the antibacterial activity of orange peel extract,and the results provide a reference for further investigation of the antibacterial activity of orange peel extract.
