摘要
为确定广东省某动物园1只患病海南坡鹿(Rucervus eldii hainanus)的致病菌并探究其主要生物学特性,采集患病坡鹿脓汁,通过细菌分离纯化、形态学观察、16S rRNA基因测序分析和生化试验鉴定对分离株进行种属鉴定;通过药敏试验和毒力基因检测对分离菌进行生物学特性分析。结果显示:该菌株菌落形态呈微弱β溶血、光滑湿润、乳白色、中间凸起且边缘整齐;革兰氏染色呈圆形革兰氏阳性短杆状菌。16S rRNA基因测序分析显示,该菌与10株化脓隐秘杆菌(Trueperella pyogenes)参考菌株同源性在98.81%以上;生化试验鉴定显示,分离菌对蔗糖、麦芽糖和葡萄糖等呈阳性,对乳糖、鼠李糖和甘露醇等呈阴性。综合以上结果确定该分离株为化脓隐秘杆菌。药敏试验结果显示,该菌对青霉素、头孢曲松和头孢克肟等抗菌药物耐药。毒力基因检测结果显示,分离株携带Plo-g2、Plo-g1、NanP、NanH、FimA、FimE和FimC七种毒力基因。研究结果可为野生动物源化脓隐秘杆菌的防治提供参考依据,为化脓隐秘杆菌的进一步研究提供素材。
In order to identify the pathogenic bacteria of a Rucervus eldii hainanus in the zoo in Guangdong Province and ex⁃plore its main biological characteristics,the pus of diseased animals was collected,and the isolates were identified by bacte⁃rial isolation and purification,morphological observation,16S rRNA gene sequencing and biochemical test.The biological characteristics of the isolated bacteria were analyzed by drug sensitivity test and virulence gene detection.The results showed that the colony morphology of the strain was weak beta hemolysis,smooth and moist,milky white,convex in the middle,and neat at the edges.Gram stain showed round gram-positive brachy rod-shaped bacteria.16S rRNA gene se⁃quencing analysis showed that the homology of this bacterium was more than 98.81%with 10 reference strains of Trueperella pyogenes.Biochemical tests showed that the isolates were positive for sucrose,maltose and glucose,but negative for lac⁃tose,rhamnose and mannitol.Based on the above results,the isolate was identified as T.pyogenes.The results of drug sen⁃sitivity test showed resistance to penicillin,ceftriaxone,cefixime and other antibacterial drugs.Virulence gene test results showed that the isolates carried seven virulence genes,including Plo-g2,Plo-g1,NanP,NanH,FimA,FimE and FimC.The results can provide reference for the prevention and control of T.pyogenes from wild animal,and provided material for further research on T.pyogenes.
