超声辅助酶法提取莲子心多糖工艺优化、结构表征及减轻过量运动诱导的骨骼肌氧化应激研究

Optimization of Ultrasound Assisted Enzymatic Extraction Process,Structural Characterization,and Alleviation of Skeletal Muscle Oxidative Stress Induced by Excessive Exercise for Plumula nelumbinis Polysaccharides

摘要 以莲子心为原料,探讨莲子心多糖的最佳提取工艺条件及结构特征,并考察其对过量运动诱导的骨骼肌氧化应激损伤的缓解作用。通过单因素、正交试验及响应面法优化超声辅助酶法提取莲子心多糖的工艺,采用高效液相色谱(High performance liquid chromatography,HPLC)、紫外光谱扫描(Ultraviolet,UV)、傅里叶变换红外光谱扫描(Fourier transform infrared spectroscopy,FT-IR)探究该多糖的单糖成分及结构特征,并利用过量运动小鼠疲劳模型观察其对骨骼肌组织中氧化应激指标的影响。结果表明,当复合酶最佳用量为纤维素酶2%、果胶酶2%和木瓜蛋白酶4%(溶液总质量的质量分数)时,超声辅助酶法提取莲子心多糖的最佳工艺条件为液料比40:1(mL/g)、超声功率180 W、酶解pH5.0、酶解温度54℃、酶解时间81 min,多糖得率为(14.94±0.31)%。经HPLC分析该多糖化合物的单糖组成包含甘露糖、鼠李糖、葡萄糖、半乳糖及阿拉伯糖,UV与FT-IR光谱均显示莲子心多糖存在多糖的特征吸收信号。动物试验结果显示,与对照组2相比,试验组1、2小鼠的超氧化物歧化酶和谷胱甘肽过氧化物酶活力明显提高(P<0.01),力竭游泳时间显著延长(P<0.05,P<0.01),上述现象与灌胃剂量呈正相关性,表明莲子心多糖能减轻过量运动诱导的骨骼肌氧化应激损伤,改善运动疲劳状态。因此研究结果可为莲子心多糖的开发与利用提供参考。 With the Plumula nelumbinis as raw materials,the optimum extraction process of polysaccharides from Plumula nelumbinis and its structural feature were explored.Meanwhile,its alleviation effect on oxidative stress injure in skeletal muscle induced by excessive exercise was investigated.The single factor test,orthogonal test and response surface method were employed to optimize the extracting process of polysaccharides from Plumula nelumbinis with ultrasonic-assisted enzymatic method,respectively.The structural properties and monosaccharide composition of polysaccharides extract from Plumula nelumbinis were explored by high performance liquid chromatography(HPLC),ultraviolet(UV)and Fourier transform infrared spectroscopy(FT-IR).Excessive exercise mice model was established to investigate its effect on oxidative stress index in skeletal muscle.The results showed that the optimum extraction conditions were as follows:liquid-solid ratio of 40:1(mL/g),ultrasonic power of 180 W,enzymatic temperature of 54℃and pH of 5.0,enzymatic time of 81 min,when the dosage of complex enzyme were as follows:2%cellulase,2%pectinase,4%papain(mass fraction of total solution weight).Under these conditions,the extraction yield of polysaccharides was(14.94±0.31)%.HPLC analysis showed that the polysaccharides from Plumula nelumbinis contained mannose,rhamnose,glucose,galactose and arabinose.Simultaneously,the spectrum of UV and FT-IR manifested that there was a polysaccharides characteristic absorption signal in the extract.The results showed that comparing with control group 2,the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in mice of experimental group 1,2 could be increased(P<0.01)and significantly prolonged the time of exhaustive swimming(P<0.05,P<0.01),which above phenomenons corresponded positively with intragastric dosage.These results indicated that the oxidative stress injure in skeletal muscle induced by excessive exercise was alleviated by polysaccharides from Plumula nelumbinis,and improved exercise fatigue in body.Therefore,this study could provide a reference for exploration and utilization of polysaccharides from Plumula nelumbinis.
