小鼠硅沉着病发生发展过程中肺部菌群的变化 被引量:1

Changes in lung microbiota during the occurrence and development of silicosis in mice

摘要 目的:分析小鼠硅沉着病发生发展过程中肺部菌群的变化。方法:将60只6周龄的SPF级C57BL/6N雄性小鼠分为对照组和SiO_(2)组,每组各30只,将50 mg/mL SiO_(2)悬液50μL(SiO_(2)组)或无菌生理盐水50μL(对照组)注入小鼠咽部。分别于染毒后第7、28和56天处死小鼠,每组取4只小鼠肺组织,实时荧光定量PCR法检测IL-6、TNF-α、COL1A1和α-SMA mRNA的表达;收集余6只小鼠的支气管肺泡灌洗液,采用16S rDNA测序法进行菌群检测。结果:与对照组比较,随时间进展,SiO_(2)组小鼠肺组织肺泡结构被破坏、炎性细胞浸润、胶原纤维沉积增加。第7、28、56天两组菌群α-多样性各指标差异无统计学意义(P>0.05),β-多样性存在明显差异(P<0.05)。KEGG和COG分析结果显示,优势菌群主要富集到氨基酸、碳水化合物等代谢通路。结论:硅沉着病的发生发展伴随着肺部菌群的失调,肺纤维化过程与氨基酸、碳水化合物等的代谢存在关联。 Aim:To observe the changes in lung microbiota during the occurrence and development of silicosis in mice.Methods:Sixty 6-week-old SPF-grade C57BL/6N male mice were allocated into control group and SiO_(2) group,with 30 mice in each group.50 mg/mL SiO_(2) suspension(for the SiO_(2) group)or sterile normal saline(for the control group)50μL was injected into the pharynx of the mice.Ten mice were sacrificed on the 7th,28th and 56th days after exposure,respectively.The lung tissues of 4 mice in each group were taken,and the expressions of IL-6,TNF-α,COL1A1 andα-SMA mRNA were detected by real-time fluorescence quantitative PCR.The bronchoalveolar lavage fluid of the remaining 6 mice was collected,and the microbiota was detected by 16S rDNA sequencing.Results:Compared with control group,as time progressed,in the lung tissues of the mice in the SiO_(2) group,there were damages to the alveolar structure,infiltration of inflammatory cells,and an increase in the deposition of collagen fibers.There was no significant difference in the indexes ofα-diversity of the microbiota between the 2 groups on the 7th,28th and 56th days(P>0.05),but there were obvious differences in theβ-diversity(P<0.05).The results of KEGG and COG analyses showed that the dominant microbiota were mainly enriched in metabolic pathways such as those of amino acids and carbohydrates.Conclusion:The occurrence and development of silicosis are accompanied by the dysregulation of the lung microbiota,and the process of pulmonary fibrosis is associated with the metabolism of amino acids,carbohydrates and so on.
