硒代蛋氨酸对猪精液常温保存效果的影响

Effects of selenomethionine on preservation of boar semen at room temperature

摘要 为了提高常温保存期间猪精子的质量,延长精子存活时间,试验采集8头杜洛克公猪的精液,在猪精液常温保存稀释液中添加不同浓度(0,0.1,1,10 mg/L)的硒代蛋氨酸(SeMet),构建过氧化氢(H_(2)O_(2))氧化损伤模型,检测常温保存第1,3,5,7,9天猪精子活力、活率、质膜完整率及顶体完整率和氧化应激条件下精子内谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量及活性氧(ROS)水平。结果表明:常温保存至第5~9天时,与未添加SeMet的A组相比,C组(添加1 mg/L SeMet)的精子活力与活率显著上升(P<0.05),同时显著提高了精子的质膜和顶体完整率(P<0.05);在氧化损伤模型中,H_(2)O_(2)的添加显著提高了精子内MDA含量和ROS水平(P<0.05),但经SeMet处理后猪精子内的GSH-Px、SOD活性显著升高(P<0.05),同时显著降低MDA含量和ROS水平(P<0.05)。说明在猪精液常温保存稀释液中添加1 mg/L的SeMet可以显著提高猪精液中抗氧化物酶的活性,从而提高猪精液品质和保存时间。 In order to improve the quality of porcine sperm and prolong survival time during room temperature preservation,semen from 8 Duroc boars was collected.Selenomethionine(SeMet)of different concentrations(0,0.1,1 and 10 mg/L)was added to the diluent of pig semen stored at room temperature and the oxidative damage model of hydrogen peroxide(H_(2)O_(2))was constructed.The motility,vitality,plasma membrane integrity and acrosome integrity of porcine sperm at 1,3,5,7,9 days after storage at room temperature and the activities of glutathione peroxidase(GSH-Px,)superoxide dismutase(SOD)and catalase(CAT),the content of malondialdehyde(MDA)and reactive oxygen species(ROS)in spermatozoa under oxidative stress conditions were detected.The results showed that the sperm vitality and motility in the C group(1 mg/L SeMet)were significantly increased(P<0.05),and the plasma membrane and the acrosome integrity of sperm were significantly enhanced(P<0.05)when stored at room temperature for 5~9 days.In the oxidative damage model,H_(2)O_(2) supplementation significantly increased the contents of MDA and ROS in the sperm(P<0.05),but the activities of GSH-Px and SOD in porcine sperm were significantly increased after SeMet treatment(P<0.05),while the contents of MDA and ROS were significantly decreased(P<0.05).The results indicated that adding 1 mg/L SeMet into diluent of pig semen stored at room temperature could significantly enhance the activities of antioxidant enzymes in pig semen,and thus improve the quality and storage time of pig semen.
