The role of endophilin Ⅰ in Remifentanil-induced hyperalgesia in rats

摘要 目的观察Endophilin I在瑞芬太尼致痛敏的大鼠海马组织中表达水平的变化,并研究其在痛觉过敏的作用。方法取12只雄性SD大鼠随机分为2组(每组6只),分为瑞芬太尼组和对照组。采用尾静脉输注瑞芬太尼的方法建立痛觉过敏模型,分别利用Von Frey及Hargreaves法检测瑞芬太尼输注后2h至72h的机械及热刺激撤足阈值变化。采用免疫印迹(Western blot)方法检测大鼠海马组织中Endophilin I、μ阿片受体和NR1的表达变化,并用免疫荧光(Immunofluorescence)观察Endophilin I在大鼠海马组织中的表达定位情况。结果在尾静脉输注瑞芬太尼后,机械及热刺激的结果均显示大鼠的痛觉阈值出现明显下降(P<0.01)。免疫荧光和免疫印迹的结果显示瑞芬太尼的输注可以上调大鼠海马组织中Endophilin I的表达(P<0.01),NR1和μ阿片受体则在免疫印迹的检测结果中分别呈现上调(P<0.01)和下调(P<0.01)的趋势。结论尾静脉输注瑞芬太尼可致大鼠痛觉过敏,使Endophilin I在大鼠海马组织中的表达出现上调,这可能是瑞芬太尼诱导的痛觉过敏的机制之一。 Objective To explore the expression of Endophilin Ⅰ in the hippocampus of rats and its role during the development of hyperalgesia induced by the infusion of remifentanil. Methods 12 male SD rats were randomly divided into 2 groups,control group and remifentanil group(6 rats for each group). Hyperalgesia was induced by intravenous infusion of remifentanil for 2 hours.And nociception was assessed by Von Frey test and Hargreaves methods from 2 hours to 72 hours after remifentanil infusion. The changes of expression of Endophilin Ⅰ,NR1 and μ opioid receptor in hippocampus was observed in immunofluorescence and western blot analysis. Results Intravenous infusion of remifentanil led to significant hyperalgesia. Immunofluorescence and Western blot results showed that the expression of Endophilin Ⅰ was significantly upregulated(P<0.01)in the hippocampus of rats of hyperalgesia induced by remifentanil infusion,and NR1 was also upregulated(P<0.01),while μopioid receptor were downregulated(P<0.01).Conclusion Intravenous infusion of remifentanil led to significant hyperalgesia and upregulated theexpression of Endophilin Ⅰ. The upregulation of Endophilin Ⅰ was in accordance with the development of hyperalgesiaplayed a role in the endocytosis of μopioid receptor,which suggested that the upregulation of Endophilin Ⅰ may be a underlying mechanism of hyperalgesia induced by remifentanil.
