孟德尔随机化法分析胃癌与足跟骨密度间的因果关系及作用机制

摘要 目的通过两样本孟德尔随机化方法,探讨胃癌(GC)和足跟骨密度(Heel bone mineral density)之间的因果关系和作用机制。方法从全基因组关联研究(GWAS)中获取足跟骨密度和胃癌的数据,采用R软件4.2.1版、方差反加权(Inverse variance weighting,IVW)和加权中位数等方法进行,用R软件环境中的“TwoSampleMR”包(0.5.7版)提供评估、测试和进行因果效应敏感性分析。与骨密度和胃癌相关的数据集来自基因表达综合(GEO)数据库。通过limma软件包筛选差异表达基因(deg)。使用Cluster Profiler和GO plot软件包进行富集分析,以验证MR分析的结果。结果MR数据分析提示了足跟骨密度与胃癌存在因果关系,通过生物信息学分析了足跟骨密度和胃癌的遗传学机制,提示其可促进胃癌的发生和发展,并为以后预防胃癌提供科学依据。结论足跟骨密度与胃癌之间存在因果关系。 Objective To investigate the causal relationship between gastric cancer(GC)and heel bone mineral density(BMD)and the mechanism of action by using the two-sample Mendelian randomization method.Methods Data on heel BMD and gastric cancer were obtained from genome-wide association studies(GWAS)using R software version 4.2.1,inverse variance weighting(IVW),and weighted median methods.The TwoSampleMR package(version 0.5.7)in the R software environment was used for evaluation,testing,and causal sensitivity analysis.Datasets related to bone mineral density and gastric cancer were sourced from the Gene Expression Omnibus(GEO)database.Differentially expressed genes(DEGs)were identified using the limma software package.Enrichment analysis was conducted using the Cluster Profiler and GO plot software packages to validate the results of the MR analysis.Results MR data analysis indicated a causal relationship between heel bone mineral density and gastric cancer.Bioinformatics analysis of the genetic mechanisms underlying heel bone mineral density and gastric cancer suggested that it may promote the occurrence and development of gastric cancer,providing a scientific basis for future gastric cancer prevention.Conclusions There is a causal relationship between heel bone mineral density and gastric cancer.
