摘要
目的探究逍遥聪耳方对肝郁脾虚型耳鸣(TLSSD)大鼠的防治作用及其初步作用机制。方法取7~8周龄、体重190~210 g的SPF级雄性Wistar大鼠42只,其中8只为空白组,34只采用复合病因法(过度疲劳、饮食失节、束缚应激共4周)诱导肝郁脾虚证,水杨酸钠(175 mg/kg)腹腔注射4周的方式诱导耳鸣。32只大鼠造模成功,按照随机数字表法将大鼠分成白组、模型组、阳性组(卡马西平15 mg/kg)、低剂量组(生药量15 g/kg)、高剂量组(生药量30 g/kg),每组8只。灌胃给药与耳鸣造模同期进行,均持续4周。于给药前后进行中医证候量化评分,采用听觉惊跳反射前刺激抑制试验(GPIAS)监测耳鸣行为学;治疗后采用实时RT-qPCR检测耳蜗肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)基因表达;液质联用法测定下丘5-羟色胺(5-HT)、多巴胺(DA)含量。结果模型组GPIAS抑制率低于空白组(P<0.05)。给药4周后,与模型组比较,高、低剂量组中医证候量化评分降低,GPIAS抑制率增高,耳蜗TNF-α、IL-6 mRNA相对表达量降低,下丘5-HT、DA含量升高(P<0.05);高剂量组下丘5-HT含量高于低剂量组(P<0.05),高、低剂量组中医证候量化评分、GPIAS抑制率、耳蜗TNF-α和IL-6 mRNA相对表达量、下丘DA含量比较,差异无统计学意义(P>0.05)。结论逍遥聪耳方对TLSSD大鼠的治疗作用可能与其调控炎症因子和神经递质、减轻神经炎症反应、影响中枢可塑性、改善情绪障碍等有关。
Objective To explore the therapeutic effect of Xiaoyao Conger Decoction on tinnitus with syndrome of tinnitus with syndrome of liver-qi stagnation and spleen deficiency(TLSSD)in rats and its preliminary mechanisms.Methods A total of 42 SPF male Wistar rats aged 7 to 8 weeks and weighing 190 to 210 g,eight of them were blank group,34 of them were selected to induce liver depression and spleen deficiency syndrome by compound etiological method(excessive fatigue,diet disorder,and restraint stress for four weeks),and tinnitus was induced by intrabitoneal injection of sodium salicylate(175 mg/kg)for four weeks.A total of 32 rats were successfully modeled,they were divided into blank group,model group,positive group(Carbamazepine 15 mg/kg),low dose group(crude drug 15 g/kg),and high dose group(crude drug 30 g/kg)by random number table method,with eight rats in each group.Drugs were orally administered for four week.The traditional Chinese medicine syndrome scores were documented before and after administration;tinnitus severity in rats was assessed by the gapprepulse inhibition of the acoustic startle reflexl(GPIAS)test for tinnitus;tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)mRNA expression in the cochlea were detected by RT-qPCR;and the contents of 5-hydroxytryptamine(5-HT)and dopamine(DA)in the inferior colliculus were determined by LC-MS/MS.Results The inhibition rate of GPIAS in model group was lower than that in blank group(P<0.05).After four weeks of administration,compared with model group,the quantification score of traditional Chinese medicine syndrome in high and low dose groups decreased,the inhibition rate of GPIAS increased,the relative expression levels of TNF-αand IL-6 mRNA in cochlear decreased,and the contents of 5-HT and DA in inferior colliculus increased(P<0.05).The content of 5-HT in the lower colliculus of high dose group was higher than that in the low dose group(P<0.05).There were no significant differences in the quantitative score of traditional Chinese medicine syndrome,the inhibition rate of GPIAS,the relative expression of TNF-αand IL-6 mRNA in cochlear and the content of DA in the lower colliculus of high dose group and low dose group(P>0.05).Conclusion The therapeutic effect of Xiaoyao Conger Decoction on TLSSD rats may be related to its ability to regulate the inflammatory factor and neurotransmitters,reduce the neuroinflammatory response,influence the central plasticity and improve the mood disorders,etc.
