三角梅自交不亲和相关基因转录组学分析

摘要 为探究三角梅(Bougainvillea spp.)孢子体自交不亲和反应过程中相关基因的互作情况,以三角梅‘伊娃夫人’(B.glabra'Mrs.Eva')自交(♀伊娃夫人×♂伊娃夫人)和杂交([♀伊娃夫人×♂小叶紫(B.glabra'Sao paulo')]授粉不同时期(30 min、3 h和24 h)的柱头为材料,进行转录组测序,比较自交和杂交授粉在基因表达水平上的差异。结果表明,授粉后3 h和24 h,参与自交授粉反应的差异表达基因数量明显多于杂交授粉反应;自交授粉反应过程中,下调基因占主导地位;杂交授粉反应过程中,上调基因占主导地位。差异表达基因GO功能注释和富集分析结果表明,自交和杂交授粉反应过程中存在部分共同生物过程。对自交授粉不亲和相关基因(24 h)进行挖掘,筛选出32个蛋白激酶类相关基因和17个泛素化相关基因,这些基因均在自交授粉反应过程中表达上调,在杂交授粉反应过程中表达不明显。研究结果丰富了三角梅自交不亲和转录组信息,可为进一步挖掘和揭示三角梅自交不亲和机制提供依据。 In order to explore interactions of related genes in process of self-incompatibility reaction of Bougainvillea spp.sporophyte,stigmas of B.glabra'Mrs.Eva'at different periods of self-pollination(♀B.glabra'Mrs.Eva'×♂B.glabra'Mrs.Eva')and cross-pollination(♀B.glabra'Mrs.Eva'×♂B.glabra'Sao paulo')(30 min,3 h and 24 h)were used as transcriptome sequencing materials to compare differences at gene expression levels between self-pollination and cross-pollination.Results showed that numbers of differentially expressed genes involved in self-pollination reaction were obviously more than those in cross-pollination reaction at 3 h and 24 h after pollination.Down-regulated genes dominated in process of self-pollination reaction,while up-regulated genes dominated in process of cross-pollination reaction.Results of GO function annotation and enrichment analysis of differentially expressed genes showed that there were some common biological processes between processes of self-pollination and cross-pollination reactions.Self-incompatibility related genes(24 h)were digged,and 32 protein kinase related genes and 17 ubiquitination related genes were screened,all of which were up-regulated in process of self-pollination reaction but not in process of cross-pollination reaction.Results of the study could enrich transcriptome information of self-incompatibility of Bougainvillea spp.and provide basis for further digging and revealing mechanisms of self-incompatibility of Bougainvillea spp..
