低频电脉冲通过RhoA/Rho激酶信号通路在流产模型子宫内的影响

Effects of low-frequency electrical pulses via the RhoA/Rho kinase signaling pathway in utero in an abortive model

摘要 目的探讨低频电脉冲通过RhoA/Rho激酶信号通路在流产模型子宫内的影响。方法选取在河北省实验动物中心购入的30只雌性大鼠和15只雄性大鼠进行交配,将未怀孕的雌性大鼠分为正常组,怀孕的雌性大鼠随机分为模型组和低频电脉冲组,正常组的大鼠在处死前未接受任何治疗,模型组用于构建人工流产模型,低频电脉冲组采用电针疗法治疗。检测并比较3组宫内膜厚度、腺体数、纤维化面积比例、E-cadherin、β-catenin、CLDN1蛋白及Rho GTP酶激活蛋白(ArhGAP1)信使RNA(mRNA)、RhoA mRNA、ROCK1 mRNA表达水平,以及RhoA、ROCK1、白细胞介素(IL)-6、肿瘤坏死因子-α(TNF-α)蛋白表达水平。结果正常组、模型组、低频电脉冲组的大鼠各有10只。低频电脉冲组子宫内膜厚度高于模型组,子宫内腺体数多于模型组,差异均有统计学意义(P<0.05)。正常组子宫内膜厚度高于模型组,子宫内膜腺体数多于模型组,差异均有统计学意义(P<0.05)。正常组和低频电脉冲组子宫内膜纤维化面积比例低于模型组,差异均有统计学意义(P<0.05)。模型组E-cadherin蛋白、β-catenin、CLDN1蛋白表达水平高于正常组,低频电脉冲组E-cadherin蛋白、β-catenin蛋白表达水平低于模型组,CLDN1蛋白表达水平高于模型组,差异均有统计学意义(P<0.05)。模型组ArhGAP1 mRNA表达水平低于正常组,RhoA mRNA和ROCK1 mRNA表达水平高于正常组,差异均有统计学意义(P<0.05)。低频电脉冲组ArhGAP1 mRNA表达水平高于模型组,RhoA mRNA、和ROCK1 mRNA表达水平低于模型组,差异均有统计学意义(P<0.05)。模型组RhoA蛋白、ROCK1蛋白、IL-6蛋白、TNF-α蛋白表达水平高于正常组和低频电脉冲组(P<0.05)。结论人工流产后大鼠给予低频电脉冲可以抑制RhoA/Rho激酶信号,减少炎症反应和子宫内膜纤维化。 Objective To investigate the effects of low frequency electrical pulses on the uterus of an abortive model through RhoA/Rho kinase signaling pathway.Methods A total of 30 female rats and 15 male rats purchased from Hebei Experimental Animal Center were selected for mating.The non-pregnant female rats were divided into normal group,and the pregnant female rats were randomly divided into model group and low frequency electric pulse group.The rats in the normal group were not given any treatment before death,and the model group was used to construct the induced abortion model.The low frequency electric pulse group was treated with electroacupuncture therapy.Endometrium thickness,number of uterine glands,endometrial fibrosis area ratio,E-cadherin,β-catenin,CLDN1 protein,Rho Gtpase activating protein(ArhGAP1)messenger RNA(mRNA),RhoA mRNA and ROCK1 mRNA and RhoA,ROCK1,interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)protein expression levels were detected and compared among the 3 groups.Results There were 10 rats in normal group,model group and low frequency electric pulse group.The endometrial thickness and the number of uterine glands in the low frequency electric pulse group were higher than those in the model group,and the differences were statistically significant(P<0.05).The endometrial thickness and number of uterine glands in normal group were higher than those in model group,and the differences were statistically significant(P<0.05).The proportion of endometrial fibrosis area ratio in normal group and low frequency electric pulse group was lower than that in model group,with statistical significance(P<0.05).The expression levels of E-cadherin protein,β-catenin protein and CLDN1 protein in model group were higher than those in normal group,the expression levels of E-cadherin protein andβ-catenin protein in low frequency electric pulse group were lower than those in model group,and the expression levels of CLDN1 protein in low frequency electric pulse group were higher than those in model group,with statistical significance(P<0.05).The expression level of ArhGAP1 mRNA in model group was lower than that in normal group,and the expression levels of RhoA mRNA and ROCK1 mRNA in model group were higher than those in normal group,with statistical significance(P<0.05).The expression levels of ArhGAP1 mRNA in low frequency electric pulse group were higher than those in model group,while the expression levels of RhoA mRNA,ROCK1 mRNA in low frequency electric pulse group were lower than those in model group,with statistical significance(P<0.05).The expression levels of RhoA protein,ROCK1 protein,IL-6 protein and TNF-αprotein in model group were higher than those in normal group and low frequency electric pulse group(P<0.05).Conclusion Low frequency electric pulse can inhibit RhoA/Rho kinase signal and reduce inflammation and endometrial fibrosis in rats after induced abortion.
