基于代谢组学分析有机肥对蒙古黄芪生长发育的影响

Metabolomics Analysis of the Effects of Organic Fertilizers on the Growth and Development of Astragalus membranaceus

摘要 以二年生蒙古黄芪为试材,采用不同浓度“元泰丰”有机肥处理及广泛靶向代谢组学技术,研究了适宜其根系生长的有机肥浓度,并对最适有机肥浓度处理(OF)与无施肥处理(CK)进行代谢物种类、含量差异分析,并探讨组间KEGG富集通路,以期为进一步对其药用价值的研究、人工栽培育种改良提供参考依据。结果表明:施用6 000 kg·hm^(-2)的有机肥可以有效提高黄芪的根长、根粗、干质量、鲜质量等农艺性状。代谢组学分析可知,在蒙古黄芪根系中共鉴定出黄酮类、酚酸类、脂质、氨基酸及其衍生物等十一大类,1 317种代谢物;多元统计分析可知,2个处理间次生代谢物差异明显,主要包括黄酮类(147)、酚酸类(32)、萜类(20)等250种物质,其中2-羟基齐墩果酸(2-hydroxyoleanolic acid)、3-羟基光甘草酚(3-hydroxyglabrol)、剑叶龙血素A(cochinchinenin A)等为主要差异物质;与CK相比,OF处理下大多差异代谢物为上调表达,且以黄酮类物质为主;同时KEGG分析发现,差异代谢物主要富集到异黄酮生物合成、黄酮和黄酮醇生物合成类和类黄酮生物合成3条黄酮类物质合成通路。综上所述,不同“元泰丰”有机肥施用下,蒙古黄芪根系生长差异显著,其中以有机肥浓度为6 000 kg·hm^(-2)的表现最佳;最适施肥浓度下,蒙古黄芪根系大多次生代谢产物呈上调表达,以黄酮类物质最为显著。 Taking biennial Astragalus membranaceus as the test material,different concentrations of‘Yuantaifeng’organic fertilizer treatment and widely targeted metabolomics techniques were used,the optimal concentration of organic fertilizer for root growth was studied,and the optimal organic fertilizer concentration treatment(OF)and no fertilizer treatment(CK)were compared by analyzing the differences in metabolite types and contents,and the KEGG enrichment pathway between groups were explored,in order to provide reference for further research on its medicinal value and artificial cultivation and breeding improvement.The results showed that the application of 6000 kg·hm^(-2) organic fertilizer could effectively improve the agronomic traits of A.membranaceus,such as root length,root diameter,dry weight,and fresh weight.Metabolomics analysis revealed that a total of 11 major categories,including flavonoids,phenolic acids,lipids,amino acids,and their derivatives,were identified in the root system of Astragalus membranaceus,with 1317 metabolites identified.Multivariate statistical analysis showed that there were significant differences in secondary metabolites between treatments,mainly including 250 substances such as flavonoids(147),phenolic acids(32),terpenes(20),etc.Among them,2-hydroxyoleanolic acid,3-hydroxyglycyrrhizic acid,and cochinchinchinanin A were the main differential substances.Compared with CK,most differential metabolites under OF treatment were upregulated and mainly composed of flavonoids.Meanwhile,KEGG analysis revealed that differential metabolites were mainly enriched in three flavonoid biosynthesis pathways,isoflavone biosynthesis,flavonoid and flavonol biosynthesis,and flavonoid biosynthesis.In summary,there were significant differences in root growth of Astragalus membranaceus under different organic fertilizer applications,with the best performance observed at an organic fertilizer concentration of 6000 kg·hm^(-2).At the optimal fertilization concentration,most of the secondary metabolites in the roots of Astragalus membranaceus were upregulated,with flavonoids being the most significant.
