尿路感染病原体即时检测微流控系统研制 被引量:1

Research and fabrication of point-of-care testing microfluidic system for pathogen of urinary tract infection

摘要 为了实现现场快速诊断,研制了一种基于离心微流控技术的尿路感染病原体即时检测(POCT)系统。该系统由离心检测芯片、选通阀进样芯片和POCT装置组成。利用比色反应的原理,可在60 min内直接用肉眼观察到检测结果。实验定量结果表明,该系统对一定浓度范围内大肠杆菌富集效率为99.8%,检测限为10~5CFU/mL。最后,通过对大肠杆菌浓度梯度为10~5~10~8 CFU/mL的4组模拟样品进行检测,记录下4组浓度对应的检测结果。将结果制成标准比色卡,便于进一步对检测结果进行分析。该系统成本低、易操作、检测快,具有一定的应用价值。 In order to achieve rapid diagnosis on site,a system which can achieve point-of-care test(POCT)of urinary tract infection pathogen is researched and fabricated based on the centrifugal microfluidic technology.The system is composed of centrifugal detection chip,selector valve injection chip and POCT device.Take advantage of the theory of colorimetric reaction,the result can be observed by naked eyes within 60 minutes.The quantitative results of experiment suggest that the device can reach a recovery rate of E.coli to 99.8%in a certain range,and the limitation of detection is 105 CFU/mL.At last,four-groups of simulattion samples with concentration gradients of 105~108 CFU/mL E.coli is detected and the testing results corresponding to the four groups concentrations are recorded.These results are translated to a standard colorimetric card for further analysis of detection result.This system has the advantages of low cost,easy to operate and fast detection,has a certain application value.
