摘要
肝脏有着独特的再生功能,同时,肝脏在人体很多重要的需要相互协调、相互制约的生物学过程中都扮演着举足轻重的角色。越来越多的证据表明,miRNA参与调节肝脏的生长、再生和代谢。因此,肝内的miRNA网络的改变与包括肝炎、肝硬化、脂肪肝、肝癌等诸多肝脏疾病有关。其中,miR-122是肝脏miRNA中含量最丰富也是肝脏生物学和疾病的最关键因子。目前,miR-122已经被证实是丙型肝炎关键宿主因子和药物靶标。这对使用直接靶向药物治疗和干扰素治疗是个有力的补充。文章综述总结了目前所了解到的miR-122在肝脏生理学和疾病中所扮演的角色,其在丙肝和乙肝中的作用以及基于miRNA的治疗肝脏疾病的方法。
As the largest organ in the human body,the liver has the particular faculty to regrow.Meanwhile,the liver plays a critical role in the biological processes that need mutual coordinations and restrictions.There is growing evidence that miRNAs are involved in regulating the growth,regeneration and metabolism of the liver.Therefore,the change of miRNAs network in the liver is associated with many liver diseases,such as hepatitis,cirrhosis,fattyliver,and liver cancer,etc.Among them,miR-122 is the most abundant miRNA expressed in the liver and a key factor of the liver biology and disease.Recently,miR-122 has been shown to be an essential host factor for hepatitis C virus infection and an antiviral target,which is complementary to the standard of care using direct-acting antivirals or interferon-based treatment.The role of miR-122 in liver physiology and disease,its role in hepatitis C and hepatitis B,and mirRNA-based approaches to the treatment of liver disease are summarized.
