摘要
研究旨在实现LMH贴壁细胞悬浮培养、稳定传代、高密度生长,并应用于血清4型禽腺病毒(FAdV-4)悬浮培养。研究进行LMH贴壁细胞低含量血清适应驯化,并进行无血清悬浮培养筛选及FAdV-4 RP-4毒株接毒盲传试验。结果显示:LMH悬浮细胞可在无血清培养基中传代第3代后,最终细胞密度达4.00×10^(6)cells/mL以上;FAdV-4连续培养至第4代,毒价可达10^(8.13)TCID_(50)/0.1 mL。研究表明,用特定培养基可使LMH贴壁细胞实现悬浮驯化,并可应用于FAdV-4稳定增殖。
The purpose of this study is to realize LMH adherent cell suspension culture,stable passage and high density growth,and apply it to serum avian adenovirus type 4(FAdV-4)suspension culture.In this study,LMH adherent cells were acclimated with low serum content,screened by serum-free suspension culture and blind transmission test of FAdV-4 RP-4 strain.The results showed that LMH suspension cells could be cultured in serum-free medium for the third generation,and the final cell density was over 4.00×10^(6)cells/mL.The toxicity of FAdV-4 could reach 10^(8.13)TCID_(50)/0.1 mL when cultured to the fourth generation.The results showed that LMH adherent cells could be domesticated in suspension with specific medium,and could be applied to the stable proliferation of FAdV-4.
