摘要
为了解人工养殖南方鲇(Silurus meridionalis)肠道菌群结构以及抗性基因的组成,采用宏基因组测序技术和生物信息学方法分析人工养殖南方鲇肠道菌群,分离验证了6株不同的细菌。结果显示,肠道菌群共注释到47个门、1425个属、4171个种,其中类志贺邻单胞菌(Plesimonas shigelloides)为优势种,3个样本中相对丰度分别为23.3%、23.4%和26.4%。KEGG分析显示,南方鲇肠道菌群的基因主要参与糖类代谢和氨基酸代谢过程。GO分析显示,南方鲇肠道菌群的基因主要参与代谢过程、细胞成分和催化反应。CARD数据库注释,鉴定出5151个抗性基因,注释到31种抗生素类型,其中有7个抗性基因未注释到具体抗生素。成功分离、培养和纯化了类志贺邻单胞菌、嗜水气单胞菌(Aeromonas hydrophila,)、维氏气单胞菌(A.veroni)、温和气单胞菌(A.sobria)、地衣芽孢杆菌(Bacillus licheniformis)和乳酸乳球菌(Lactococcus lactis)6种细菌。本研究结果为南方鲇肠道菌群结构提供初步分析,并统计了抗性基因携带情况,以期为南方鲇人工养殖病害防治提供参考数据。
To understand intestinal microflora structure and composition of resistance genes of cultured southern catfish Silurus meridionalis,six different species of bacteria were selected from metagenomic sequencing technology and bioinformatics methods for isolation.The results showed that the intestinal flora was annotated to 47 phyla,1425 genera,and 4171 species,with dominant species Plesimonas shigelloides,and the relative abundances in 3 samples were 23.3%,23.4%,and 26.4%,respectively.KEGG analysis showed that the genes of southern catfish intestinal flora were primarily involved in carbohydrate metabolism and amino acid metabolism.GO analysis showed that the genes in gut flora of southern catfish were mainly involved in metabolic processes,cellular component and catalytic reactions.Moreover,resistance gene annotation results showed 5151 resistance genes were identified,and annotated to 31 antibiotic types,without annotation to specific antibiotics in 7 resistance genes.Six species of bacteria were successfully isolated,cultured and purified:Plesiomonas shigelloides,Aeromonas hydrophila,Aeromonas veroni,Aeromonas sobria,Bacillus licheniformis,Lactococcus lactis.The findings provided preliminary analysis of intestinal microflora structure and preliminary statistics of resistance gene carrying in southern catfish,aiming to provide reference data for the prevention and control of diseases in southern catfish culture.
