摘要
本文建立了电位滴定测定面粉添加剂葡萄糖氧化酶活度的新方法。葡萄糖氧化酶催化氧化葡萄糖产生H_(2)O_(2),高价铈(Ce^(4+))与H_(2)O_(2)发生2∶1的定量反应,当用Ce^(4+)标准溶液滴定H_(2)O_(2)时,H_(2)O_(2)生成量直接体现了葡萄糖氧化酶活度,考察了滴定条件,系统分析了方法的准确度和精密性。结果表明,葡萄糖氧化酶活度在0.00~3.40 U/mL范围内与Ce^(4+)标准溶液滴定体积呈线性关系:∆V(Ce^(4+))=6.061X(U/mL)-0.0223,相关系数为0.9988,方法的最低检出量为4.94×10^(-3)U/mL。自配葡萄糖氧化酶工作液作测试液,平行6次测定,相对标准偏差RSD<5%。
A new method for the potentiometric titration determination of glucose oxidase activity in flour additives was established.Glucose oxidase catalyzes the oxidation of glucose to produce H_(2)O_(2),Cerium(IV)ion(Ce^(4+))undergone a 2∶1 quantitative reaction with H_(2)O_(2),where when titrating H_(2)O_(2)with a standard Ce^(4+)solution,the amount of H_(2)O_(2)generated directly reflected the glucose oxidase activity.The titration conditions were investigated,and the accuracy and precision of the method were systematically analyzed.The results showed that the glucose oxidase activity in the range of 0.00 to 3.40 U/mL was linearly related to the titration volume of the standard Ce^(4+)solution:∆V(Ce^(4+))=6.061X(U/mL)-0.0223,with a correlation coefficient of 0.9988.The method’s limit of detection was 4.94×10^(-3)U/mL.Using a self-prepared glucose oxidase working solution as the test solution,six parallel measurements were conducted with a relative standard deviation(RSD)of less than 5%.
