KMT2C在胃癌中的表达特征和功能分析

摘要 目的:探讨组蛋白赖氨酸N-甲基转移酶2C[lysine(k)-specific methyltransferase 2C,KMT2C]在胃癌中的表达、作用及临床意义。方法:采用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)检测KMT2C在150例胃癌组织及相应的正常组织中的表达水平。χ^(2)检验、Kaplan-Meier生存曲线以及Cox回归模型分析KMT2C表达与胃癌患者临床病理特征及预后的相关性。CCK-8实验、集落形成实验、伤口愈合实验和Transwell迁移侵袭实验检测敲低KMT2C对胃癌细胞增殖、迁移和侵袭能力的影响。结果:KMT2C mRNA在98例胃癌组织中的表达高于癌旁正常组织。相关性分析结果显示,KMT2C mRNA表达与TNM分期(P=0.032)和T分期(P=0.044)显著相关。多因素Cox回归分析结果显示,KMT2C mRNA表达水平(HR=1.693,95%CI:1.034~2.774,P=0.037)以及TNM分期(HR=1.903,95%CI:1.127~3.212,P=0.016)是影响胃癌预后的独立危险因素。KMT2C高表达患者的总生存期显著低于低表达患者(P<0.05)。此外,敲低KMT2C表达可以抑制胃癌细胞SGC-7901的增殖、迁移和侵袭能力。结论:KMT2C作为促癌基因在胃癌中高表达并与不良预后相关。KMT2C可能作为胃癌治疗的潜在靶点。 Objective:To investigate the expression,function,and clinical significance of histone lysine(k)-specific methyltr-ansferase 2C(KMT2C)in gastric cancer.Methods:Quantitative real-time PCR was used to detect the expression of KMT2C in 150 gastric cancer tissues and corresponding normal tissues.Chi-square test,Kaplan-Meier survival analysis,and Cox regression model were employed to analyze the correlation between KMT2C expression and clinicopathological features and prognosis in gastric cancer patients.CCK-8 experiment,colony formation experiment,wound healing experiment and Transwell migration and invasion experiment were performed to evaluate the impact of KMT2C knockdown on the proliferation,migration,and invasion abilities of gastric cancer cells.Results:The expression of KMT2C mRNA in 98 gastric cancer tissues was higher than that in normal tissues.Correlation analysis showed that KMT2C mRNA expression was significantly correlated with TNM stage(P=0.032)and T stage(P=0.044).Multivariate Cox regression analysis demonstrated that KMT2C mRNA expression level(HR=1.693,95%CI:1.034-2.774,P=0.037)and TNM stage(HR=1.903,95%CI:1.127-3.212,P=0.016)were independent risk factors affecting prognosis of gastric cancer.Overall survival was significantly lower in patients with high KMT2C expression compared to those with low expression(P<0.05).Furthermore,knockdown of KMT2C expression inhibited proliferation,migration,and invasion abilities of gastric cancer cells(SGC-7901).Conclusion:KMT2C is highly expressed as an oncogene in gastric cancer and is associated with poor prognosis.KMT2C may be a potential target for the treatment of gastric cancer.
