2022年~2023年上半年河北省猪繁殖与呼吸综合征病毒的遗传变异分析 被引量:12

Genetic diversity and phylogenetic analysis of PRRSV in Hebei Province from 2022 to the first half of 2023

摘要 为了解当前河北省猪繁殖与呼吸综合征病毒(PRRSV)的流行及流行株的遗传变异情况,本研究利用荧光定量PCR方法对2022年~2023年上半年于河北省不同规模养猪场采集的229份疑似PRRSV感染猪的组织病料样品进行检测,结果显示,PRRSV阳性率为17.9%(41/229)。对鉴定的15株PRRSV ORF5基因进行测序、遗传进化分析、以及潜在的N-糖基化位点分析。结果显示,15株PRRSV ORF5基因序列的同源性为81.8%~99.8%,氨基酸序列的同源性为81.1%~100.0%;15株PRRSV均属于PRRSV-2型,其中的10株属于谱系1(Lineage 1),1株属于Lineage 5,4株属于Lineage 8;属于Lineage 5的HB-HD-2株为一株类RespPRRS MLV疫苗株。部分病毒株的GP5蛋白氨基酸序列毒力相关位点出现R13Q、R151I、R151K突变,属于Lineage 1的10株PRRSV GP5蛋白在诱骗表位出现V27A和V29A突变,在其它中和表位区及高变区也出现了不同程度的变异;糖基化位点分析结果显示,GP5蛋白在aa44~aa46、aa51~aa53位N-糖基化位点相对保守,在aa30~aa32、aa33~aa35、aa34~aa36和aa57~aa59发生了不同程度的突变。以上结果表明,当前河北省PRRSV流行株复杂多样,Lineage 1 PRRSV(NADC30-Like、NADC34-Like)正成为优势病毒株,猪繁殖与呼吸综合征(PRRS)防控形势仍较为严峻。本研究为掌握当前河北省PRRSV的遗传变异情况,以及PRRS疫苗开发和防控提供了新数据和信息。 To understand the current epidemic situation of porcine reproductive and respiratory syndrome virus(PRRSV)and the genetic variation of the circulating strain in Hebei Province,229 tissue samples from pigs with suspected PRRSV infection symptoms were collected from farms with different sizes in Hebei Province from 2022 to the first half of 2023.PRRSV was detected by real-time fluorescence-based quantitative PCR.Results showed that 41 of 229 materials were PRRSV-positive,with a positive rate of 17.9%.The sequence,genetic evolution,and potential N-glycosylation sites of the ORF5 gene from 15 isolated strains were analyzed.Results showed that the nucleotide and amino acid sequence homology of 15 obtained PRRSV ORF5 genes were 81.8%to 99.8%and 81.1%to 100.0%,respectively.All the 15 PRRSV belonged to PRRSV-2,of which 10 strains belonged to Lineage 1,1 strain in Lineage 5,and 4 strains in Lineage 8;the HB-HD-2 strain,which belonged to Lineage 5 was a Resp PRRS MLV-like vaccine strain.Virulence-related sites in GP5 amino acid in the some of the isolated strains showed R13Q,R151I,and R151K mutations,and 10 strains that belonged to Lineage 1 showed V27A and V29A mutations at the decoy epitope.Different degrees of gene variation also appeared in other neutral epitope regions and hypervariable regions.The N-glycosylation sites of GP5 at aa44-aa46 and aa51-aa53 were relatively conserved,however,there are mutations at aa30-aa32,aa33-aa35,aa34-aa36,and aa57-aa59.These results indicated that the current epidemic strains of PRRSV in Hebei Province were complex and diversified.Lineage 1 PRRSV(NADC30-Like,NADC34-Like)began to be a prevalent Lineage.Better prevention and control measures for porcine reproductive and respiratory syndrome(PRRS)were urgently needed.Results of this study enriched the epidemiological data and current genetic variation on PRRSV in Hebei Province.It provided new reference for the development of new vaccines and for the prevention and control of PRRS.
