清热利湿健脾方对溃疡性结肠炎大鼠肠道菌群、免疫炎性反应及内质网应激的影响 被引量:1

The Effect of Qingre Lishi Jianpi Formula(清热利湿健脾方) on Gut Microbiota,Immune Inflammatory Response,and Endoplasmic Reticulum Stress in Rats with Ulcerative Colitis

摘要 目的 探讨清热利湿健脾方对溃疡性结肠炎(UC)大鼠肠道菌群、免疫炎性反应及内质网应激的影响。方法 雄性SD大鼠SPF级40只,随机分为4组,每组10只。制备UC大鼠模型。低剂量组大鼠给予1.42 g/kg清热利湿健脾方汤灌胃,持续14 d;高剂量组大鼠给予5.68 g/kg清热利湿健脾方灌胃,持续14 d;模型组和正常组大鼠灌胃氯化钠溶液等剂量,持续14d。比较各组大鼠DAI评分,肠道菌群,免疫炎性因子,ATF4、CHOP和GRP78蛋白表达情况。结果 与正常组比较,其余各组UC大鼠DAI评分、大肠杆菌和肠球菌相对表达量、IL-6、IL-17、ATF4、CHOP和GRP78蛋白表达灰度值升高,而乳酸杆菌和双歧杆菌相对表达量、IL-10水平降低(P<0.05);与模型组比较,低剂量组和高剂量组UC大鼠DAI评分、大肠杆菌和肠球菌相对表达量、IL-6、IL-17、ATF4、CHOP和GRP78蛋白表达灰度值降低,而乳酸杆菌和双歧杆菌相对表达量、IL-10水平升高(P<0.05);与低剂量组比较,高剂量组UC大鼠乳酸杆菌和双歧杆菌相对表达量、IL-10水平降低,而乳酸杆菌和双歧杆菌相对表达量、IL-10水平升高(P<0.05)。结论 清热利湿健脾方可改善UC大鼠肠道菌群,减轻免疫炎性反应,下调ATF4、CHOP及GRP78蛋白表达。 Objective To explore the effects of Qingre Lishi Jianpi Formula(清热利湿健脾方) on gut microbiota,immune inflammatory response,and endoplasmic reticulum stress in rats with ulcerative colitis(UC).Method Forty male SD rats of SPF grade divided four groups,each group 10 rats.Prepare UC rat model.Low dose group rats were given 1.42 g/kg Qingre Lishi Jianpi Decoction by gavage for 14 days;High dose group rats were given 5.68 g/kg of Qingre Lishi Jianpi Formula by gavage for 14 days;the model group and normal group rats were gavaged with physiological saline at the same dosage for 14 days.Compare the DAI scores,gut microbiota,immune inflammatory factors,and expression of ATF4,CHOP,and GRP78 proteins in each group of rats.Result Compared normal group,the DAI score,relative expression levels of Escherichia coli and Enterococcus,IL-6,IL-17,ATF4,CHOP,and GRP78 protein expression gray values of UC rats in other groups increased,while Lactobacillus and Bifidobacterium relative expression,and IL-10 levels decreased(P<0.05);Compared model group,the low-dose and high-dose groups showed a decrease in DAI scores,relative expression levels of Escherichia coli and Enterococcus,and grayscale values of IL-6,IL-17,ATF4,CHOP,and GRP78 protein expression in UC rats,while Lactobacillus and Bifidobacterium relative expression,and IL-10 levels increased(P<0.05);compared to the low-dose group,the high-dose group showed a decrease in the relative expression levels and IL-10 levels of Lactobacillus and Bifidobacterium in UC rats,while the relative expression levels and IL-10 levels of Lactobacillus and Bifidobacterium increased(P<0.05).Conclusion The Qingre Lishi Jianpi formula can improve the intestinal microbiota of UC rats,alleviate immune inflammatory response,and downregulate the expression of ATF4,CHOP,and GRP78 proteins.
