摘要
为研究解整合素-金属蛋白酶9(ADAM9)与藏鸡胚胎低氧适应性的关系,试验对藏鸡和茶花鸡在低氧(O_(2),13%)和常氧(O_(2),21%)孵化条件下胚胎尿囊绒毛膜(CAM)组织ADAM9基因mRNA和蛋白表达量进行检测,同时检测了在不同氧气浓度培养条件下鸡胚成纤维细胞(DF-1)中ADAM9基因的mRNA表达变化和ADAM9基因启动子区活性。结果显示:不论藏鸡还是茶花鸡,除第18天外,在低氧孵化下CAM组织中ADAM9的表达量显著高于常氧孵化同一时期(P<0.05),并且在低氧条件下培养的DF-1细胞中ADAM9基因mRNA表达量在36 h时显著高于常氧条件(P<0.05)。结果表明低氧能刺激藏鸡胚CAM组织中ADAM9基因的表达,从而使其胚胎能更好地适应低氧环境。
In order to study the relationship between ADAM metallopeptidase domain 9(ADAM9)and the hypoxic adaptabil⁃ity of Tibetan chicken embryos,the mRNA and protein expression of ADAM9 gene in embryonic chorioallantoic membrane(CAM)tissues of Tibetan chicken and Chahua chicken under hypoxic(O_(2),13%)and normoxic(O_(2),21%)incubation conditions were de⁃tected.The expression of ADAM9 gene in chicken embryonic fibroblast cell(DF-1)cultured in different oxygen concentration was compared and ADAM9 gene promoter activity was identified.The results showed that the expression of ADAM9 in CAM tissues un⁃der hypoxic incubation was significantly higher than that under normoxic incubation at the same time(P<0.05),except for the 18th day.The expression of ADAM9 mRNA in DF-1 cells cultured under hypoxia condition was significantly higher than that un⁃der normoxia condition at 36 h(P<0.05).The results indicated that hypoxia could stimulate the expression of ADAM9 gene in CAM tissue of Tibetan chicken embryos,so that the embryos could adapt to the hypoxic environment better.
