摘要
目的探讨YAP在增生性瘢痕中的表达及作用。方法自2018年9月至2020年9月,从北部战区总医院烧伤整形科采集了4例患者身上4份增生性瘢痕组织样本,4例皮瓣手术患者剩余的正常皮肤组织样本,通过蛋白免疫印迹实验检测组织中YAP表达水平。获取原代增生性瘢痕成纤维细胞,分成3组,分别为空白组、对照组(si-NC)和si-YAP组,转染后24 h蛋白免疫印迹实验检测YAP和I型胶原蛋白表达水平,CCK-8实验在转染后0、1、2、3、4 d检测细胞增殖水平,Transwell实验检测细胞迁移水平。结果YAP在增生性瘢痕组织中表达上调,高于正常组织中(P<0.05)。Si-NC转染并不影响细胞中蛋白的表达、细胞的增殖和迁移能力。与对照组相比,si-YAP组显著下调增生性瘢痕成纤维细胞中YAP和Ⅰ型胶原蛋白的表达水平(P<0.05)。转染后0、1、2、3、4 d时,YAP沉默均能够显著下调细胞的增殖水平(P<0.05)。沉默YAP 24 h后细胞的迁移能力显著下调(P<0.05)。结论YAP在增生性瘢痕组织中表达上调,能够促进Ⅰ型胶原蛋白表达和增生性瘢痕成纤维细胞的增殖和迁移水平。
Objective To investigate the expression and role of YAP in hypertrophic scar.Methods From September 2018 to September 2020,Department of Burn and Plastic Surgery of General Hospital of Northern Theater Command collected 4 samples of hypertrophic scar tissue from 4 patients and the remaining normal skin tissue samples from 4 patients undergoing flap surgery.The expression of YAP in hypertrophic scar tissue and normal skin tissue was detected by Western blot.Primary hypertrophic scar fibroblasts were obtained and divided into three groups:blank control group,si-NC group and si-YAP group.The expression of YAP and Collagen I was detected by Western blot at 24 h after transfection,and then the cell proliferation was detected by CCK-8 at 0,1,2,3,4 d after transfection.Transwell assay was used to detect cell migration.Results The expression of YAP was up-regulated in hypertrophic scar tissues,which was higher than that in normal tissues(P<0.05).Si-NC transfection did not affect protein expression,cell proliferation and migration.Compared with si-NC(control group),si-YAP significantly decreased the expression of YAP and Collagen I in hypertrophic scar fibroblasts(P<0.05).At 0,1,2,3,4 d after transfection,YAP silencing could significantly down regulate cell proliferation(P<0.05).After silencing YAP for 24 h,the migration ability of cells was significantly down regulated(P<0.05).Conclusion YAP is up-regulated in hypertrophic scar tissue,which can promote the expression of Collagen I,proliferation and migration of hypertrophic scar fibroblasts.
