摘要
种子含水量密切影响种子活力。蒙古栎种子新采收后含水量较高,不耐贮藏。为探究蒙古栎种子含水量对种子耐藏性的影响,将不同含水量蒙古栎种子分别贮藏在低温(4℃)和室温(20℃)条件下,并检测其生理生化变化,评价其耐藏性。研究以当年采收的蒙古栎种子为试验对象,利用硅胶干燥法分别控制种子含水量为50%(采后初始含水量)、40%、30%和20%。在贮藏期间,定期测定种子萌发率和主要生理生化指标。结果表明:蒙古栎种子最适贮藏含水量为30%~40%。室温条件下,种子含水量为30%耐藏性较好,最长贮藏时间为56 d,并在此期间维持种子萌发率达30.10%以上。低温条件下,种子含水量为40%时的贮藏效果最好。40%含水量种子贮藏84 d后萌发率仍可达45.45%。蒙古栎种子贮藏过程中,可溶性糖在前期快速合成,而后稳定下降。同一贮藏条件下,降低种子含水量能有效提升可溶性蛋白含量,减少丙二醛产生,并促进游离脯氨酸增加。低温贮藏条件下丙二醛和游离脯氨酸积累普遍低于室温贮藏。综合结果表明:降低种子初始含水量,在室温和低温贮藏下均可增强蒙古栎种子的耐藏性,低温贮藏整体优于室温贮藏。研究结果为蒙古栎种子合理存储提供了科学依据。
The seed water content closely affects seed vigor.The seed of Quercus mongolica have high water content after new harvest and are not resistant to storage.In order to investigate the effect of seed water content of Q.mongolica on seed storage tolerance,the seeds with different water content were stored at low temperature(4℃)and room temperature(20℃),and their physiological and biochemical changes were detected to evaluate their storage tolerance.In this study,the seeds harvested in the current year were used as test subjects,and the seed water content was set as 50%(initial postharvest water content),40%,30% and 20%(silica gel drying method),respectively.During the storage,the germination rate and main physiological and biochemical indexes of seeds were measured regularly.The results showed that the optimum storage seed water content of Q.mongolica was 30%-40%.At room temperature,the seeds had good storage tolerance when the water content was 30%.The longest storage time was 56 days,and the seed germination rate was more than 30.10% during this period.At low temperature,the seed water content was 40%,and the storage effect was the best.The germination rate of seeds with 40% water content could reach 45.45% after 84 days of storage.During the storage of Q.mongolica seeds,soluble sugars were synthesized rapidly in the early stage and then steadily decreased.Under the same storage conditions,reducing seed water content could effectively increase the content of soluble protein,reduce the production of malondialdehyde(MDA),and promote the increase of free proline.The accumulation of MDA and free proline in seeds under low temperature storage was generally lower than that under room temperature storage.These results showed that the storage tolerance of Q.mongolica seeds could be enhanced by reducing the initial seed water content under both room temperature and low temperature,and low temperature storage was better than room temperature storage.The study provided scientific basis for rational seed storage of Q.mongolica.
