摘要
TIFY转录因子参与调节植物的生长和发育,同时参与植物应激反应,在植物生长中具有重要调控功能。为探索TIFY基因家族成员在大豆胞囊线虫病(Soybean Cyst Nematode,SCN)3号生理小种抗性反应中的作用,选择抗病品种东农L-10和感病品种黑农37进行SCN 3号生理小种胁迫处理,在转录组测序数据中分析筛选到多个与抗性相关的TIFY转录因子,并对其进行生物信息学预测和分析。结果表明:从转录组数据中鉴定和筛选到23个与SCN 3号生理小种相关的差异表达TIFY转录因子,其中18个转录因子上调表达,5个转录因子下调表达,TIFY基因在大豆的根、茎、叶均存在一定表达量,在相同的大豆胞囊线虫胁迫下,抗病品种中TIFY表达量要高于感病品种。不同TIFY转录因子之间的理化性质存在较大差异。23个转录因子均属于亲水性蛋白,多数为酸性等电点。亚细胞定位预测结果显示,TIFY转录因子发挥重要作用的位置依次为细胞核、细胞质、线粒体。蛋白质二级结构中氨基酸序列以无规则卷曲为主要组成,α-螺旋次之,β-折叠和β-转角则散布于蛋白质序列中。基因分布在大豆的10条染色体上,磷酸化位点丝氨酸偏多,启动子中存在很多响应激素应答和胁迫相关的顺式作用元件,说明TIFY基因所编码的蛋白中部分为响应胁迫的蛋白。研究结果说明筛选到的23个与SCN 3号生理小种相关的TIFY转录因子可为进一步了解大豆TIFY基因的功能提供重要参考。
TIFY transcription factors are involved in the regulation of plant growth and development,as well as plant stress response,and play an important role in plant growth regulation.In order to explore the role of TIFY gene family members in the resistance of soybean cyst nematode(Soybean Cyst Nematode,SCN)race 3,in this study,resistant variety Dongnong L-10 and susceptible variety Heinong 37 were treated with SCN3 race stress.Several TIFY transcription factors related to resistance were screened from transcriptome sequencing data,and bioinformatics prediction and analysis were carried out.The results showed that 23 differentially expressed TIFY transcription factors related to race SCN3 were identified and screened from transcriptome data,of which 18 were up-regulated and 5 down-regulated.TIFY gene was expressed in roots,stems and leaves.Under the same stress of soybean cyst nematode,the expression of TIFY in resistant varieties was higher than that in susceptible varieties.There were great differences in physical and chemical properties among different TIFY transcription factors.23 transcription factors belong to hydrophilic proteins,most of them were acidic isoelectric points.The prediction results of subcellular localization showed that TIFY transcription factors play an important role in nucleus,cytoplasm and mitochondria.In the protein secondary structure,the amino acid sequence was mainly composed of irregular curl,followed byα-helix,β-fold andβ-rotation scattered in the protein sequence.Genes were distributed on 10 chromosomes of soybean,there was more serine in the phosphorylation site.There were many cis-acting elements in the promoter that respond to hormone response and stress.There were interaction proteins in the protein encoded by TIFY gene.The results showed that 23 TIFY transcription factors related to race SCN3 could provide an important reference for further understanding the function of TIFY gene in soybean.
