摘要
转录是细菌基因表达调控的关键步骤,也是重要的抗生素靶标。探究转录调控过程中转录因子与RNA聚合酶(RNA polymerase,RNAP)、脱氧核糖核酸(deoxyribonucleic acid,DNA)以及核糖核酸(ribonucleic acid,RNA)之间的相互作用以及这些蛋白-核酸复合物的结构对于深入理解原核转录调控的分子机制至关重要。随着直接电子探测器和图像处理算法的进步,冷冻电镜技术已经成为转录调控机制研究的重要手段。本文综述了近年来利用冷冻电镜完成的重要原核转录调控机制研究,并展望冷冻电镜在原核转录领域未来的应用前景。
Transcription is a key step in the regulation of bacterial gene expression and an important antibiotic target.Exploring the interactions between transcription factors and RNA polymerase,DNA and RNA,as well as the structures of these protein⁃nucleic acid complexes in the process of transcription regulation is crucial for a deep understanding of the molecular mechanism of prokaryotic transcription regulation.With the advancement of direct electron detectors and image processing algorithms,Cryo⁃electron microscopy has become an important tool for the study of transcriptional regulation mechanism.This article reviews the important research on the regulation mechanism of prokaryotic transcription using Cryo⁃electron microscopy in recent years and looks forward to the future application prospects of Cryo⁃electron microscopy in the field of prokaryotic transcription.
