颗粒细胞线粒体DNA拷贝数用于评估胚胎发育潜能的临床价值 被引量:1

Clinical Application Research of Evaluating Embryonic Development Potential with Granulosa Cell Mitochondrial DNA Copy Numbers

摘要 目的 探究颗粒细胞线粒体DNA拷贝数用于评估胚胎发育潜能的临床应用。方法 选取2020年1月-2021年12月本院辅助生殖技术助孕的新型卵裂期胚胎总计90个,选择数字聚合酶链式反应(PCR)检测胚胎培养液中颗粒细胞线粒体DNA拷贝数(mtDNA),依据胚胎评分分为高分组(n=70)、低分组(n=20),对比基础内分泌条件、妊娠结局等的差异。结果 高分组在FSH、LH、Hcg日内膜厚度、mtDNA拷贝数高于低分组,E2、P低于低分组,差异有统计学意义(P<0.05)。高分组妊娠成功率高于低分组,差异有统计学意义(P<0.05)。结论 胚胎培养液中对mtDNA拷贝数的获取是进行胚胎质量评估的一项重要手段,与形态学评分相结合能评价胚胎质量,当mtDNA拷贝数越高则胚胎质量越好,有助于辅助生殖技术中选择最优胚胎植入,提高妊娠率。 Objective To explore clinical application of granulosa cell mitochondrial DNA copy numbers in evaluation of embryonic development potential.Methods The paper chose 90 new cleavage stage embryos transferred by assisted reproductive technology from January 2020 to December 2021 in our hospital,and detectd mitochondrial DNA copy numbers in granulosa cells in embryo culture medium with digital Polymerase chain reaction(PCR),and divided them into high group(n=70)and low group(n=20)according to embryo scores.Difference in basic endocrine conditions and pregnancy outcomes was compared.Results Endometrial thickness and mtDNA copy numbers on FSH,LH,and Hcg days in high group were higher than low group,while E2 and P were lower in low group;success rate of pregnancy in high group was 57.14%,which was higher than 30.00%in low group,difference was statistically significant(P<0.05).Conclusion Obtaining mtDNA copy number in embryo culture medium is an important means to evaluate embryo quality.Combining with morphological scores,it can evaluate embryo quality.When mtDNA copy number is higher,embryo quality is better,which is helpful to chose the best embryo implantation in assisted reproduction technology and improve pregnancy rate.
