茯苓酸调节Lyn/Syk信号通路对哮喘大鼠气道炎症的影响 被引量:2

Effects of pachymic acid on airway inflammation in asthmatic rats by regulating Lyn/Syk signal pathway

摘要 目的探讨茯苓酸对支气管哮喘大鼠气道炎症及酪氨酸蛋白激酶(Lyn)/脾酪氨酸激酶(Syk)信号通路的影响及其机制。方法建立支气管哮喘大鼠模型,将大鼠分为对照组、哮喘组、Lyn抑制剂PP2组、茯苓酸5 mg/kg组及茯苓酸10 mg/kg组,观察大鼠肺组织病理学变化,并进行炎症评分。ELISA法检测支气管肺泡灌洗液(BALF)与血清TNF-α、IFN-γ、IL-4水平,Giemsa染色法检测BALF炎症细胞,qRT-PCR法检测肺组织Lyn、Syk、STAT3 mRNA表达水平,Western blot检测肺组织Lyn、Syk、p-STAT3蛋白表达。结果哮喘组大鼠支气管黏膜水肿,管腔内容物增多,微血管渗漏,并伴有大量炎性细胞浸润。与对照组比较,哮喘组大鼠支气管炎症评分、BALF中炎性细胞数目、血清及BALF中TNF-α、IL-4水平及肺组织Lyn、Syk、STAT3mRNA及蛋白表达水平显著升高,BALF中IFN-γ水平显著降低(P<0.05);与哮喘组比较,PP2组、茯苓酸5 mg/kg组与茯苓酸10 mg/kg组的上述改变均得以减轻(P<0.05),且茯苓酸10 mg/kg组比茯苓酸5 mg/kg组效果为明显(P<0.05)。结论茯苓酸可抑制支气管哮喘大鼠气道炎症反应,其机制可能与抑制Lyn/Syk信号通路有关。 To investigate the effects of pachymic acid on airway inflammation and tyrosine protein kinase Lyn/spleen tyrosine kinase(Syk)signal pathway in rats with bronchial asthma and its mechanism.A rat model of bronchial asthma was established,and the rats were grouped into asthma group,Lyn inhibitor PP2 group,pachymic acid 5 mg/kg group and pachymic acid 10 mg/kg group,with normal rats as control group.The pathological changes in the lung tissue of the rats were observed and the inflammatory score was evaluated;the levels of TNF-α,IFN-γand IL-4 in bronchoalveolar lavage fluid(BALF)and serum were detected by ELISA;the BALF inflammatory cells were detected by Giemsa staining;the mRNA expression levels of Lyn,Syk and STAT3 in lung tissue were detected by qRT-PCR,and the expression of Lyn,Syk and p-STAT3 proteins in lung tissue were detected by Western blotting.In the asthma group,the bronchial mucosa was edema,the contents of the lumen increased,the microvessel leaked,and a large number of inflammatory cells infiltrated.Furthermore,as compared with the control group,the score of bronchitis,the number of inflammatory cells in BALF,the levels of TNF-αand IL-4 in serum and BALF,as well as the mRNA and protein expression of Lyn,Syk,STAT3 in lung tissue of asthma group were obviously increased,while the level of IFN-γin BALF was obviously decreased(P<0.05).In the PP2 group,the 5 mg/kg pachymic acid group and the 10 mg/kg pachymic acid group,all those changes mentioned in asthma group were reversed,and 10 mg/kg pachymic acid was more effective,as compared with 5 mg/kg pachymic acid(P<0.05).In conclusion,pachymic acid can inhibit airway inflammation in rats with bronchial asthma,and its mechanism may be related to the inhibition of Lyn/Syk signal pathway.
