北山林场不同林分表层土壤化学性质与细菌群落差异分析

Differences of Chemical Properties and Bacterial Communities in Topsoil of Different Stands in Beishan Forest Farm

摘要 为探究青海互助北山林场内典型纯林表层土壤化学性质与细菌群落差异,选取林区内山杨、云杉、青扦和白桦四种林分为研究对象,用铝盒与环刀在28 m×28 m树样方内沿对角线每隔10 m分别取林下土壤样本,采用PacBio平台的16S三代测序技术对土壤细菌16S rRNA的V1~V9区域进行高通量测序和生物信息学分析。结果表明:不同林分间除有机质外其他化学性质均存在显著性差异(P<0.05)。四种林分聚类后共得到2531个OTU;细菌α多样性结果表明云杉的Shannon指数、Chao1指数和ACE指数显著低于白桦、青扦、山杨;β多样性分析表明四种林分表层土壤细菌在结构组成上相对稳定且存在差异;四种林分表层土壤中优势菌门为变形菌门(Proteobacteria),优势菌属为不明酸杆菌(unidentified_Acidobacteria)。通过对本地区典型纯林表层土壤化学性质与细菌群落进行初步研究,可对青海互助森林土壤的组成以及树种改造提供数据参考。 In order to explore the differences of soil chemical properties and bacterial community of typical pure forest in Huzhu Beishan Forest Farm,Qinghai Province,in this experiment,four stands of Populus davidiana,Picea asperata,Picea wilsonii and Betula platyphylla were selected as the research objects.Soil samples were collected from 28 m×28 m tree quadrats with aluminum box and ring knife at 10 m intervals along the diagonal.16S third generation sequencing technology of pacbio platform was used for high-throughput sequencing and bioinformatics analysis of V1~V9 region of 16S rRNA of soil bacteria.The results showed that there were significant differences in chemical properties except organic matter among different stands(P<0.05).A total of 2531 OTUs were obtained from the four stands;the results of bacterial α diversity showed that Shannon index,Chaul index and ACE index of Picea asperata were significantly lower than that of Betula platyphylla,Picea wilsoni and Populus davidiana;the analysis of β-diversity showed that the structure of bacteria in the topsoil of the four stands was relatively stable and different;Proteobacteria was the dominant phylum and unidentified was the dominant genus Acidobacteria.The preliminary study on the chemical properties and bacterial community of typical pure forest surface soil in this region can provide data reference for the composition of forest soil and tree species transformation in this region.
