紫丁香EST-SSR标记在木犀科植物中通用性及亲缘关系分析 被引量:1

Analysis of the Transferability and Genetic Relationship of EST-SSR Markers of Syringa oblata in Oleaceae

摘要 木犀科植物作为广泛应用的树种,利用分子标记技术探讨其植物之间的亲缘关系,可以为木犀科植物资源的有效利用提供理论基础。本研究基于70对紫丁香EST-SSR多态性引物在29种木犀科植物中进行通用性分析并探究其亲缘关系。结果表明,70对引物在木犀科不同属植物间的通用性比率为23.8%~76.5%,平均45.4%。其中,11对引物在所有样本中的通用性和多态性比例为100%,共扩增得到114个等位基因,平均等位变异数为10.4;多态信息含量PIC的范围为0.473~0.884,平均为0.728,表明29份木犀科植物具有较高的遗传多样性。聚类结果表明,供试材料的遗传相似系数为0.19~0.94,在遗传相似系数0.69处,可分为4大类,其中素馨属、连翘属、雪柳属和流苏树属聚为第Ⅰ类,白蜡属聚为第Ⅱ类,女贞属聚为第Ⅲ类,丁香属聚为第Ⅳ类。第Ⅰ类又可分为4个小组,素馨属、连翘属、雪柳属及流苏树属分别单独成组。11对木犀科通用引物,应用在与紫丁香亲缘关系越近的植物中,通用性比率越高;分子标记水平的木犀科植物间的亲缘关系与现有植物分类结果一致。本研究筛选出11对木犀科通用引物,为木犀科植物在分子水平上的植物分类及植物资源的利用提供了有益参考。 Oleaceae plants are widely used as tree species.To explore the genetic relationship between Oleaceae plants can provide a theoretical basis of plant resources effectively based on molecular marker technology.The genetic relationship of 29 Oleaceae plants was analyzed by the universal primers selected from 70 polymorphic EST-SSRs.The results showed that the amplification rate was 23.8%~76.5%,with an average of 45.4%.Among them,11 pairs of primer could be amplified in all samples,114 alleles were obtained,with an average of 10.4 for each marker;PIC ranged from 0.473 to 0.884,with an average of 0.728,indicating that 29 Oleaceae plants had high genetic diversity.The genetic similarity coefficient of the tested materials ranged from 0.19 to 0.94.At the genetic similarity coefficient of 0.69.The plant materials could be divided into four categories,in which the genera of Jasminum,Forsythia,Fontanesia and Chionanthus were clustered into ClassⅠ,Fraxinus into ClassⅡ,Ligustrum into ClassⅢ,and Syringa into ClassⅣ.ClassⅠcould be further divided into four groups,and the Jasminum,Forsythia,Fontanesia and Chionanthus were grouped separately.11 pairs of universal primers were obtained,and the closer the genetic relationship of plants with Syringa,the higher universal ratio of EST-SSR markers in this species;the genetic relationship among Oleaceae at the molecular marker level wass consistent with existing plant classification results.These results could provide beneficial reference for identifying plants of Oleaceae family and utilization of plant resources at the molecular level.
