闪式提取无花果叶总黄酮及其抗氧化活性研究 被引量:5

Study on extraction of total flavonoids from Ficus carica L.leaves and its antioxidant activity by flash extraction

摘要 试验旨在研究闪式提取无花果叶总黄酮及其抗氧化活性。试验以无花果叶为原料,使用闪氏提取方法,以乙醇为溶剂,分别以提取时间、乙醇体积分数和液料比为单因素,测定在相应条件下总黄酮的最佳提取率,在此基础上利用响应面法对提取工艺进行优化。结果显示,响应面优化的提取工艺参数为提取时间87 s、乙醇体积分数67%、液料比87 mL/g。在此条件下,实际总黄酮提取率为3.43%,与预测值3.41%基本一致。无花果叶总黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、超氧阴离子自由基、羟自由基均有良好的清除能力,IC50值分别为0.019、0.017、0.076 g/L。研究表明,无花果叶总黄酮在体外具有良好的抗氧化能力。 The experiment was to study the extraction of total flavonoids from Ficus carica L.leaves by flash and its antioxidant activity.Ficus carica L.leaves were used as raw materials in the experiment,the method of flash extraction was used,ethanol was used as solvent,and the extraction time,ethanol volume fraction and liquid to material ratio were used as single factors to determine the optimal extraction rate of total flavonoids under corresponding conditions.On the basis of single factor experiment,response surface methodology was used to optimize the extraction process.The result showed that the final extraction of process parameters were extraction time 87 s,ethanol concentration 67%,liquid-solid ratio 87 mL/g.Under these conditions,the actual extraction rate was 3.43%,which were consistent basically with their predicted values(3.41%).The total flavones of Ficus carica L.leaves obtained had good scavenging capacity for DPPH free radicals,superoxide anion free radicals and hydroxyl free radicals,and the IC50 values were 0.019,0.017 and 0.076 g/L,respectively.The study indicates that the Ficus carica L.has good antioxidant capacity in vitro.
