摘要
该文基于过氧化钙(CaO_(2))与硫酸反应生成过氧化氢(H_(2)O_(2)),H_(2)O_(2)在甲烷氧化菌素(methanobactin,Mb)模拟酶的催化下,以KI为增敏剂,可快速氧化季胺[4,4’-(对二甲氨基)-二苯基甲烷]显色,测定溶液在462 nm处的吸光度,建立快速、简便的模拟酶催化显色法检测小麦粉中CaO_(2)。研究铜合甲烷氧化菌素(Mb-Cu)浓度、季胺用量、KI用量、反应温度、pH值以及反应时间对显色的影响。结果表明,Mb-Cu浓度4.5×10^(-5)mol/L、季胺用量60 μL,、KI用量12 μL、反应温度50℃、反应时间10 min时,CaO_(2)浓度在0~10 mg/L时与462 nm处吸光度呈线性关系,线性方程为y=0.078 1x+0.019 6,相关系数R~2=0.997 6。该方法检出限为1.82×10^(-2)mg/L(相当于2.25 mg/kg小麦粉),平均加标回收率为99.21%~100.35%,相对标准偏差为0.31%~1.22%,该方法快速、准确,可用于检测小麦粉中CaO_(2)。
In this work,as calcium peroxide (CaO_2) can react with sulfuric acid to produce hydrogen peroxide (H_2O_2) and H_2O_(2) can cause rapid oxidation and chromogenesis in quaternary amine [4,4'- (p-dimethylamino) -diphenylmethane] by using KI as the sensitizer and methanobactin (Mb) mimic enzyme as the catalyst,a rapid and simple mimic enzyme-catalyzed colorimetry for the determination of CaO_(2) in wheat flour was established after the absorbance can be detected at 462 nm.The effects of Mb-Cu concentration,quaternary amine content,KI content,reaction temperature,pH value and reaction time on chromogenic reaction were investigated.The results showed that when the concentration of Mb-Cu was 4.5×10~ (-5) mol/L,quaternary amine content was 60 μL,KI content was 12 μL,the reaction temperature was 50 ℃,and the detection time was 10 min,CaO_(2) concentration at 0-10 mg/L had a linear correlation with the absorbance at 462 nm,and the linear equation was y=0.078 1x+0.019 6,with a correlation coefficient (R~2) of 0.997 6.The limit of detection (LOD) of this method was 1.82×10~ (-2) mg/L (equivalent to 2.25 mg/kg wheat flour),the average recovery rate for spiked standards was 99.21%-100.35% and the relative standard deviation (RSD) was 0.31%-1.22%.This method is rapid and accurate,and thus it can be used for the determination of CaO_(2) in wheat flour.
