摘要
对东方蜜蜂微孢子虫RCDP 42基因进行分子克隆,并通过生物信息学解析RCDP 42蛋白的分子特性,进而对东方蜜蜂微孢子虫和其他物种的RCDP 42蛋白进行保守基序和结构域预测及分析。结果表明,成功克隆到东方蜜蜂微孢子虫RCDP 42基因的CDS区,RCDP 42基因在孢子中真实表达,含有540个核苷酸,可编码179个氨基酸;RCDP 42蛋白的分子质量约为20.3 ku,分子式为C_(915)H_(1416)N_(230)O_(274)S_(10),脂溶系数为86.03,等电点为6.57;包含的亲水氨基酸数量多于疏水氨基酸,但不含信号肽与跨膜结构域,说明RCDP 42可能为亲水性蛋白、胞内蛋白和非跨膜蛋白,可能包含16个磷酸化位点、63个α-螺旋、43条延长链、11个β-转角及62个无规则卷曲。在东方蜜蜂微孢子虫、蚱蜢脑炎微孢子虫和肠脑炎微孢子虫的RCDP 42蛋白中均鉴定到3个保守基序(motif 1、motif 2和motif 3)与1个结构域(Rho)。
RCDP 42 gene in Nosema ceranae was cloned using molecular biology approach,and the molecular characteristics of RCDP 42 protein was then analyzed using bioinformatics,followed by the prediction and investigation of conserved motifs and struc-tural domains within RCDP 42 proteins in N.ceranae and other species.The results showed that the coding sequence(CDS)region in RCDP 42 was successfully cloned,and RCDP 42 was truly expressed in N.ceranae spore;RCDP 42 contained 540 nucleotides,encoding 179 amino acids;the molecular formula of RCDP 42 protein was C_(915) H_(1416) N_(230) O_(274) S_(10),and its molecular weight,lipid sol-ubility coefficient,and isoelectric point were approximately 20.3 ku,86.03 and 6.57,respectively;RCDP 42 contained more hydro-philic amino acids than the hydrophobic ones but no signal peptide and transmembrane domain,suggesting that RCDP 42 may be hy-drophilic protein,intracellular protein and non-transmembrane protein;RCDP 42 may contain 16 phosphorylation sites,63α-heli-xes,43 extension chains,11β-turns and 62 random coils;additionally,3 conserved motifs(motif 1,motif 2 and motif 3)as well as 1 structural domain(Rho)were identified in RCDP 42 proteins in N.ceranae,Encephalitozoon romaleae and Encephalitozoon in-testinalis.These findings clarified the molecular characteristics of N.ceranae RCDP 42 gene and laid a foundation for further function-al investigation.
