摘要
为探究农村垃圾表面附着微生物的群落结构及潜在危害,选取贵州铜仁市某农村垃圾堆放场为研究对象,分析了不同常见农村垃圾的形貌及红外光谱,并利用宏基因组学技术手段分析其表面附着微生物及潜在病原体的群落结构与毒力因子.结果表明:不同垃圾的表面形貌和化学组成差异明显,垃圾表面不同程度的破损和粗糙可能有利于微生物的附着.垃圾表面附着的微生物分属222门、4460属,主要以假单胞菌属(Pseudomonas)、紫色杆菌属(Janthinobacterium)、贪噬菌属(Variovorax)为主;附着的病毒共注释28个科,以长尾噬菌体科(Siphoviridae)、肌尾噬菌体科(Myoviridae)、虹彩病毒科(Iridoviridae)为主.β多样性结果显示,垃圾附着微生物群落因材质不同具有较大的差异性.泡沫箱(M2)和易拉罐(M3)表面的病原细菌丰度值最高.垃圾表面潜在病原体主要为铜绿假单胞菌(Pseudomonas aeruginosa)、大肠杆菌(Escherichia coli)、肠道沙门氏菌(Salmonella enterica)及无脊椎虹彩病毒25(Invertebrate iridovirus 25)等,其潜在健康风险需引起重视.垃圾附着微生物群落的毒力因子主要涉及铁吸收系统和粘附,潜在病原体群落与多种毒力因子显著相关,致病途径较为多样.
To explore the community structure and potential hazards of microorganisms attached to the surface of rural garbage,different kinds of common garbage samples were collected from a typical rural garbage dump sited in Tongren City,Guizhou Province.The morphology and infrared spectrum of the six garbage samples were detected.Metagenomic technique was introduced to analyze the community structure and virulence factors of microorganisms and pathogens on the garbage surface.The results showed that the surface morphology and chemical composition of the samples were significantly different,while the varying degrees of breakage and roughness might be beneficial for microorganisms to adhere to the refuse surface.The microorganisms attached to the surface of sampled garbage belonged to 222 phyla and 4460 genera,mainly with Pseudomonas,Janthinobacterium,and Variovorax.A total of 28 families were annotated with the attached viruses,mainly abundant in Siphoviridae,Myoviridae and Iridoviridae.The results ofβdiversity showed that the microbial communities attached to the surface of garbage samples had great difference with different materials.The highest abundance of pathogenic bacteria were found in foam boxes(M2)and cans(M3).The main potential pathogens on the surface of the sampled garbage were Pseudomonas aeruginosa,Escherichia coli,Salmonella enterica and Invertebrate iridovirus 25,etc,where the potential health risks should be paid more attention to.The virulence factors of the microbial community attached to the garbage samples mainly involved iron absorption system and adhesion.And the potential pathogens were significantly related to multiple virulence factors,which might suggest the relatively diverse pathogenic pathways.
