摘要
云斑天牛(Batocera horsfieldi)是一种高度危险的林业蛀干害虫,为了探明云斑天牛的寄主适应机制,本试验以分别取食细香核桃(Juglans sigillata cv.‘Xixiang’)、娘青核桃(J.sigillata cv.‘Niangqing’)和龙佳核桃(J.sigillata cv.‘Niangqing’×J.sigillata cv.‘Yangbidapao’)枝条的云斑天牛成虫为材料,采用传统微生物分离培养与16S rDNA序列分析相结合的方法对云斑天牛成虫肠道可培养细菌进行分离鉴定,研究3种核桃寄主对云斑天牛成虫肠道可培养细菌多样性的影响。结果表明,从取食3种核桃的云斑天牛成虫肠道中共分离出19株细菌,经鉴定归类于2个门(变形菌门、厚壁菌门)9个属[拉乌尔菌属(Raoultella)、肠球菌属(Enterococcus)、沙雷氏菌属(Serratia)、肠杆菌属(Enterobacter)、乳球菌属(Lactococcus)、克雷伯氏菌属(Klebsiella)、泛菌属(Pantoea)、Gibbsiella菌属和假单胞菌属(Pseudomonas)]。Gibbsiella属为取食细香核桃后分离出的特有菌株;乳球菌属为取食娘青核桃后分离出的特有菌株;拉乌尔菌属、肠球菌属和克雷伯氏菌属为共有菌属。取食3种核桃后的云斑天牛成虫肠道可培养细菌的组成和多样性存在差异,说明寄主植物能够对云斑天牛的肠道微生物产生影响。
Batocera horsfieldi is a highly dangerous forestry drywall pest.In order to explore its host ad⁃aptation mechanism,the adult B.horsfieldi fed on branches of three species of walnut trees,namely Juglans sigillata cv.‘Xixiang’,J.sigillata cv.‘Niangqing’and J.sigillata cv.‘Niangqing’×J.sigillata cv.‘Yang⁃bidapao’,was used as material in this experiment.The culturable bacteria from the gut of adult B.horsfieldi were isolated and identified by using traditional microbial isolation and culture method combined with 16S rD⁃NA sequence analysis,and the effects of the three species of walnut hosts on the diversity of these culturable bacteria were studied.The results showed that a total of 19 strains of bacteria were isolated from the gut of a⁃dult B.horsfieldi fed on the three species of walnuts,belonging to 2 phyla(Proteobacteria and Firmicutes)and 9 genera(Raoultella,Enterococcus,Serratia,Enterobacter,Lactococcus,Klebsiella,Pantoea,Gibbsiella and Pseudomonas).Gibbsiella was identified as a specific strain isolated from B.horsfieldi individuals fed on J.sigillata cv.‘Xixiang’,while Pseudomonas was the specific strain isolated from those fed on J.sigillata cv.‘Niangqing’.Raoultella,Enterococcus and Klebsiella were identified as common strains.The composition and diversity of culturable gut bacteria in B.horsfieldi varied depending on the host plants,indicating that host plants could affect the gut microbiota of B.horsfieldi.
