基于气相色谱-串联质谱指纹图谱与化学模式识别法鉴定不同品种药用红景天 被引量:5

Identification of Different Varieties of Medicinal Rhodiola Based on GC-MS/MS Fingerprint and Chemical Pattern Recognition

摘要 目的基于气相色谱-串联质谱(GC-MS/MS)指纹图谱与化学模式识别相结合的方法对5种常见药用红景天进行比较研究。方法收集具有质量标准的5种药用红景天,采用GC-MS/MS对5种药用红景天进行差异性成分鉴定,建立各品种化合物GC-MS/MS数据库,并建立GC-MS/MS指纹图谱进行相似度评价;采用ChemPattern2017先进化学计量学系统解决方案软件对5种药用红景天建立化学模式识别法。结果分别建立5种药用红景天的GC-MS/MS指纹图谱并进行相似度评价;建立的5种药用红景天的化合物GC-MS/MS数据库可以为红景天品种鉴别提供技术支撑;采用分类模式识别分析法测试2批较细碎性状难以辨识的未知红景天,结果1批为长鞭红景天,1批为狭叶红景天;经正交偏最小二乘法分析,样品可明显分为5组,其中大花红天素、酪醇和红景天苷的VIP值>0.9,为主要差异性成分,其中大花红天素为大花红景天的独有成分;经聚类分析后分为两大类,大花红景天单独聚为一类,其余4种红景天聚为一大类,唐古红景天和长鞭红景天可单独聚为一小类,部分狭叶红景天和高山红景天混聚为一类,表明大花红景天与其余4种红景天差异特征明显,狭叶红景天和高山红景天存在一定的相似性。结论建立的方法可有效区分5种药用红景天,为5种药用红景天的质量控制、质量评价和鉴定提供了参考。 Objective Fivecommon medicinal Rhodiola were compared and studied based on gas chromatography-tandem triple quadrupole mass spectrometry(GC-MS/MS)fingerprint and chemical pattern recognition.Methods Five medicinal Rhodiola with quality standards were collected to identify the differential components of them by GC-MS/MS.The GC-MS/MS database of each compound was established,and the GC-MS/MS fingerprint was established for similarity evaluation;ChemPattern 2017 advanced stoichiometric system solution software was used to develop a chemical pattern recognition method for five medicinal rhodiola.Results The GC-MS/MS fingerprints of five medicinal Rhodiola were established and the similarity was evaluated.The GC-MS/MS compounds database of five medicinal Rhodiola was established to provide technical support for the identification of Rhodiola varieties in the future.Two batches of unknown Rhodiola rosea with unrecognizable traits were tested by classification pattern recognition analysis,and the results were that the first batch was Rhodiola fastigiata(Hook.f.et Thoms.)S.H.Fu and the other batch was Rhodiola kirilowii(Regel)Maxim;By using orthogonal partial least squares analysis,the samples could be divided into five groups,among which the VIPs value of crenulatin,tyrosol and salidroside were greater than 0.9,which were the main differentiating components,and crenulatin was the unique component of Rhodiola crenulata(Hook.f.et.Thoms.)H.Ohba;After clustering analysis,it was divided into two categories:Rhodiola renulate(Hook.f.et.Thoms.)H.Ohba was grouped into one category,the other four species were grouped into one category,Rhodiola tangutica(Maxim.)S.H.Fu,and Rhodiola fastigiata(Hook.f.et.Thoms.)S.H.Fu were grouped into one small category,and some Rhodiola kirilowii(Regel)Maxim.and Rhodiola sachalinensis A.Bor.were grouped together into one category.The results showed that Rhodiola crenulata(Hook.f.et.Thoms.)H.Ohba differed significantly from the other four species,and there were some similarities between Rhodiola kirilowii(Regel)Maxim.and Rhodiola sachalinensis A.Bor.Conclusion The established method can effectively distinguish five medicinal Rhodiola,and provide a reference for quality control,quality evaluation and identification of five medicinal Rhodiola.
