二甲双胍的研究趋势与展望

摘要 目的:通过文本挖掘探索上市药物的生命周期、不同时期的研究概况、治疗用途变化、研究热点,探究未来药物在新领域的再利用潜力。方法:以二甲双胍研究相关文本为例,针对目前医药领域文本挖掘研究方法存在的不足,采用对应分析、聚类分析和统计学的方法对数据进行降维处理。结果:揭示了二甲双胍研究发展特征及原因,发现二甲双胍在治疗肥胖症、心脑血管疾病、多囊卵巢综合征、抗癌、消炎、调节肠道微生物组和延缓衰老等方面具有老药新用价值,并且预测出二甲双胍可能在治疗阿尔茨海默病、神经退行性疾病和雄性激素脱发等方面具有药物再利用潜力;同时发现AMP活化蛋白激酶(AMPK)降糖机制的研究将是其他降糖类药物研发所参考借鉴的重点和热点。结论:实验结果证明,2000年前后是二甲双胍相关研究进入快速发展阶段的转折时期,其在多个领域都具有药物再利用价值的潜力,并预测了未来其可能存在的药物再利用价值。同时发现AMPK降糖机制是二甲双胍具有持久生命力的重要保证。 Objective Through text mining,the life cycle of drugs,the research overview in different periods,changing therapeutic uses of drugs,research hotspots and the potential for future repurposing in new areas were explored.Methods Taking the text related to metformin as an example,the data were dimensionally reduced using correspondence analysis,cluster analysis and statistics to address the shortcomings of current text mining research methods in the pharmaceutical field.Results The development characteristics and reasons of metformin were revealed,and it was found that metformin had the value of repurposing in the treatment of obesity,cardiovascular and cerebrovascular diseases,polycystic ovary syndrome,anti-cancer treatment,anti-inflammatory treatment,regulation of gut microbiome and anti-aging treatment,and it was predicted that metformin drugs may have the potential of repurposing in the treatment of Alzheimer’s disease,neurodegenerative diseases and androgenic alopecia,etc.It was also found that the study of AMPK hypoglycemic mechanism would be the focus and hot spot for the reference of other hypoglycemic drug development.Conclusion The experimental results showed that around the the year of 2000 was a turning point in the rapid development of metformin drug-related research and it had the potential value of drug repurposing in many fields,and its potential value of drug repurposing in the future was predicted.AMPK hypoglycemic mechanism was also found to be an important guarantee for the enduring vitality of metformin drugs.
