氨磷汀与香菇多糖及细胞因子联合应用对辐射损伤小鼠的保护作用 被引量:4

Synergistic Protection of WR-2721 Combined with Lentinan and Cytokine on Radiation-Injured Mice

摘要 目的探讨氨磷汀(amfostine,别名WR-2721)与香菇多糖(Lentian,LNT)、重组人粒细胞集落刺激因子(recombinant human granulocyte colony-stimulating factor,rhG-CSF)、重组人白介素-11(recombinant human Iinterleukin-11,rhIL-11)联合应用对辐射损伤小鼠的协同保护作用,为急性放射损伤的临床治疗提供一种新的治疗方案。方法取70只ICR小鼠,分为7组,分别为对照组、模型组、5个用药组,除对照组外,其他组用60Co-γ射线1次照射,剂量率为0.8 Gy·min^(-1),累积剂量为5.0 Gy。测定小鼠照前第3天、照后第3、7及14天的血常规,照后14 d处死小鼠,测定小鼠睾丸指数、骨髓DNA、骨髓有核细胞数、小鼠精子总数及睾酮和雌二醇含量。结果WR-272与LNT、rhIL-11、rhG-CSF联合应用在白细胞、红细胞、血红蛋白指标中有促进恢复的作用(P<0.05或P<0.01),小鼠睾丸指数、骨髓DNA含量、骨髓有核细胞数及小鼠精子总数均有恢复作用。结论WR-2721与LNT、rhIL-11、rhG-CSF联合应用具有较好的协同保护作用,是急性放射损伤治疗的一种较好的用药方案。 OBJECTIVE To investigate the synergistic protection of amfostine(WR-2721)combined with Lentian(LNT),granu-locyte colony-stimulating factor(rhG-CSF)and linterleukin-11(rhIL-11)on radiation-injured mice,and provide a new therapeutic scheme for acute radiation injury.METHODS Seventy ICR mice were divided into seven groups,including control group,model group and five drug groups.Except the control group,the other groups were iradiated withCo-ray at a dose rate of o.8 Cy·min^(-1) and a cumulative dose of 5.0 Gy.Routine blood tests were determined at 3 d before irradiation,3,7 and 14 d after irradiation,and the mice were sacrificed at 14 d after irradiation.Testicular index,bone marrow DNA,bone marrow nucleated cells,sperm count,testos-terone and estradiol contents of mice were determined.RESULTS The drug treatment could promote the recovery of white blood cell(WBC),red blood cell(RBC)and hemoglobin(P<0.05 or P<0.01),and the testicular index,bone marrow DNA content,bone marrow nucleated cell number and sperm count of mice.CONCLUSION WR-2721 combined with LNT,rhG-CSF and rh IL-11 has a good synergistic protection,which is a good drug regimen for the treatment of acute radiation injury.
