高效液相色谱—串联质谱法测定蜂蜜中的林可霉素残留

摘要 目的采用固相萃取技术,建立了蜂蜜中林可霉素残留的高效液相色谱-串联质谱(UPLC-MS/MS)的检测方法。方法样品以三羟甲基氨基甲烷(Tris)缓冲液提取,固相萃取柱净化,用正交设计优化提取和净化条件,采用高效液相色谱-质谱联用进行测定。结果在1.0~80μg/L范围内,线性关系良好,在3个添加水平下,林可霉素回收率为94.4%~105.0%,相对标准偏差为3.6%~7.6%方法检出限和定量限分别为0.2ug/kg,0.7ug/kg。结论该方法准确、灵敏度高,适合于蜂蜜中的林可霉素残留量的测定。 Objective A high performance liquid chromatography-mass spectrometry(UPLC-MS/MS)method was developed for the determination of lincomycin residues in honey by solid phase extraction.Methods The sample was extracted with tris buffer,purified by waters oasis column and the orthogonal design was used to optimize the extraction and purification conditions,the target compound was detected by liquid chromatography-mass spectrometer.Results The calibration curve showed good linearity in the concentration range of 1.0~80.0μg/L.The average recoveries of the lincomycin ranged from 94.4%to 105.0%with the relative standard deviations of 3.6%~7.6%the detection limit and quantitative limit of the method are 0.2ug/kg and 0.7ug/kg,respectively.Conclusion The method was found accurate and sensitive with the advantages of high throughput,which is suitable for the determination of lincomycin residues in honey.
