摘要
目的:观察母婴分离产后抑郁大鼠前额叶皮质和海马5-羟色胺(5-HT)及其代谢产物5-羟吲哚乙酸(5-HIAA)的含量,探讨不同剂量参芪解郁方的干预作用。方法:60只SD孕鼠,随机分为正常组、模型组、中药高剂量组、中药中剂量组、中药低剂量组和西药组,采用母婴分离法制备产后抑郁大鼠,灌胃两周后检测各组大鼠前额叶皮质、海马5-HT及其代谢产物5-HIAA含量。结果:模型组强迫游泳不动时间较正常组显著增加(P<0.01),蔗糖水消耗量、旷场实验水平得分和垂直得分均较正常组显著降低(P<0.01)。中药高、中、低剂量组和西药组强迫游泳不动时间较模型组显著降低(P<0.01);中药高、中、低剂量组和西药组蔗糖水消耗量、旷场实验水平得分较模型组均显著升高(P<0.01,P<0.05);中药高、中剂量组和西药组旷场实验垂直得分较模型组均显著升高(P<0.01)。模型组前额叶皮质、海马5-HT及其代谢产物5-HIAA含量较正常组均显著降低(P<0.01)。中药高、中、低剂量组和西药组前额叶皮质、海马5-HT及其代谢产物5-HIAA含量较模型组均升高(P<0.01,P<0.05)。结论:参芪解郁方通过调节产后抑郁大鼠单胺类神经递质及其代谢产物水平发挥抗抑郁作用。
Objective: To observe the contents of 5-hydroxytryptamine(5-HT) and its metabolite 5-hydroxyindolenacetic acid(5-HIAA) in prefrontal cortex and hippocampus of postpartum depression rats separated from mother and infant,and to explore the intervention effect of different doses of Shenqi Jieyu Formula. Methods: Sixty SD pregnant rats were randomly divided into normal group,model group,high dose group,medium dose group,low dose group and western medicine group. Postpartum depression rats were prepared by mother-infant separation method. After intragastric administration,the behavior of rats was evaluated and the contents of 5-HT and its metabolite 5-HIAA in prefrontal cortex and hippocampus of each group were detected.Results: The forced swim immobility time in the model group was significantly increased compared with that in the normal group(P<0.01),and the sucrose water consumption,the open field test level score,and the vertical score were all significantly decreased compared with the normal group(P<0.01). The immobility time of forced swimming in the Chinese medicine high-,medium-,and low-dose groups and the western medicine group was significantly decreased compared with that in the model group(P<0.01);sucrose water consumption and open field test level scores in the high-,medium-,and low-dose Chinese medicine groups and the western medicine group were increased compared with those in the model group(P<0.01,P<0.05). The vertical scores in the open field test in the high-,medium-dose groups,and western medicine group were significantly increased compared with those in the model group(P<0.01). The contents of 5-HT and its metabolite 5-HIAA in the prefrontal cortex and hippocampus in the model group were significantly lower than those in the normal group(P<0.01). The contents of 5-HT and its metabolite 5-HIAA in the prefrontal cortex and hippocampus in the high-,medium-and low-dose groups and the western medicine group were all higher than those in the model group(P<0.01,P<0.05). Conclusion: Shenqi Jieyu Formula exerts antidepressant effects by regulating the levels of monoamine neurotransmitters and their metabolites in postpartum depressed rats.
