摘要
目的分析肺炎克雷伯菌的喹诺酮类耐药株gyrA基因的耐药机制。方法收集经Phoenix 100全自动细菌鉴定系统鉴定分离的肺炎克雷伯菌的喹诺酮类耐药株43株,采用聚合酶链反应(PCR)方法进行gyrA基因检测,扩增产物纯化后测序并进行序列分析。结果43株肺炎克雷伯菌的喹诺酮类耐药株均发生gyrA基因突变,gyrA基因的PCR扩增均得到目的片段,扩增产物长度为441 bp。结论肺炎克雷伯菌的喹诺酮类耐药株gyrA基因突变非常普遍,监测其突变频率及特点对研究喹诺酮类耐药机制和指导临床合理用药非常必要。
Objective To analyze the drug resistance mechanism of GyrA gene of quinolone-resistant Klebsiella pneumoniae strains. Methods 43 quinolone-resistant Klebsiella pneumoniae strains identified and isolated by Phoenix 100 system were collected and detected by polymerase chain reaction(PCR) for GyrA gene,and the amplified products were purified and sequenced for sequence analysis. Results All 43 quinoloneresistant Klebsiella pneumoniae strains were found to have GyrA gene mutation. PCR amplification of the GyrA gene yielded the target fragment, and the length of the amplified product was 441 bp. Conclusion GyrA gene of quinolone-resistant Klebsiella pneumoniae strains is very common. Monitoring the mutation frequency and characteristics is very important for the study of the drug resistance mechanism of quinolone and the guidance of clinical rational drug use.
