摘要
目的 研究温肾潜阳汤对高血压肾损害大鼠的保护作用及可能机制。方法 将24只高血压肾损害大鼠随机分为3组:模型组、西药组、中药组;8只WKY大鼠作为空白对照组。西药组给予缬沙坦按8 mg/(kg·d)灌胃,中药组给予16g/(kg·d)温肾潜阳汤煎剂灌胃,空白对照组和模型组给予等体积生理盐水灌胃,每天1次,连续给药8周。检测各组大鼠的血压、Cr、BUN、m-ALB、U-TP、TGF-β1、IL-10等指标水平及肾组织病理改变。结果给药8周后,与模型组比较,中药组、西药组的收缩压、舒张压均显著降低;中药组收缩压低于西药组(P<0.05);西药组舒张压低于中药组(P<0.05)。与模型组比较,中药组、西药组的Cr、BUN、m-ALB、U-TP、TGF-β1均显著降低(P<0.05);中药组、西药组的IL-10显著升高。与模型组比较,中药组、西药组的肾脏病理损伤明显减轻。结论 温肾潜阳汤具有降低高血压肾损害大鼠血压,减少蛋白尿,保护肾功能的作用,其作用机制可能与下调TGF-β1,上调IL-10表达相关。
Objective To study the protective effect of Wenshen Qianyang Decoction(温肾潜阳汤)on rats with hypertensive renal damage and its possible mechanism.Methods Twenty-four rats with hypertensive renal damage were randomly divided into 3 groups:model group,Western medicine group,and traditional Chinese medicine group;8 WKY rats were used as blank control group.The Western medicine group was given valsartan 8 mg/(kg·d)by gavage,the traditional Chinese medicine group was given 16 g/(kg·d)Wenshen Qianyang Decoction by gavage,the blank control group and the model group were given an equal volume of normal saline by gavage,once a day,for 8 weeks.Results After 8 weeks of administration,compared with the model group,the systolic blood pressure and diastolic blood pressure of the traditional Chinese medicine group and the Western medicine group were significantly reduced;the systolic blood pressure of the traditional Chinese medicine group was lower than that of the Western medicine group(P<0.05);the diastolic blood pressure of the Western medicine group was lower than that of the traditional Chinese medicine group(P<0.05)).Compared with the model group,Cr,BUN,m-ALB,U-TP,TGF-β1 in the traditional Chinese medicine group and the western medicine group were significantly reduced(P<0.05);while the IL-10 of the Chinese medicine group and the Western medicine group were significantly increased.Compared with the model group,the kidney pathological damage in the traditional Chinese medicine group and the Western medicine group was significantly reduced.Conclusion Wenshen Qianyang Decoction has the effects of lowering blood pressure,reducing proteinuria and protecting renal function in rats with hypertensive renal damage.Its mechanism may be related to down-regulation of TGF-β1 expression and up-regulation of IL-10 expression.
