摘要
目的:探究黄连素对前列腺癌细胞增殖、凋亡能力及Wnt/β-链蛋白(Wnt/β-catenin)信号通路的影响。方法:选取生长良好的前列腺癌细胞株DU145,随机分为对照组与黄连素低、中、高剂量组,分别对各组细胞进行干预。对比细胞增殖、凋亡、迁移、侵袭情况,检测细胞Wnt/β-catenin通路表达量。结果:黄连素低、中、高剂量组各时间点细胞增殖率低于对照组,凋亡率高于对照组,差异有统计学意义(P<0.05);黄连素高剂量组细胞增殖率低于黄连素低、中剂量组,凋亡率高于黄连素低、中剂量组,差异有统计学意义(P<0.05)。黄连素低、中、高剂量组细胞侵袭细胞数、迁移细胞数低于对照组,差异有统计学意义(P<0.05);黄连素高剂量组细胞侵袭细胞数、迁移细胞数低于黄连素低、中剂量组,差异有统计学意义(P<0.05)。黄连素低、中、高剂量组细胞Bax、Caspase3、Caspase9表达高于对照组,Bcl-2表达低于对照组,差异有统计学意义(P<0.05);黄连素高剂量组细胞Bax、Caspase3、Caspase9表达高于黄连素低、中剂量组,Bcl-2表达低于黄连素低、中剂量组,差异有统计学意义(P<0.05)。黄连素低、中、高剂量组细胞Wnt、β-catenin、血管内皮生长因子(VEGF)蛋白表达量低于对照组,差异有统计学意义(P<0.05);黄连素高剂量组细胞Wnt、β-catenin、VEGF蛋白表达量低于低、中剂量组,差异有统计学意义(P<0.05)。结论:黄连素干预可促进前列癌细胞凋亡,其机制可能与调控Wnt/β-catenin信号通路相关。
Objective:To explore the effect of berberine on the proliferation and apoptosis of prostate cancer cells and Wnt/β-catenin signaling pathway.Methods:The well-grown prostate cancer cell line DU145 was selected and randomly divided into control group and low,medium and high dose groups of berberine,and the interventions were performed on each group of cells respectively.The cell proliferation,apoptosis,migration and invasion were compared,and the expression of Wnt/β-catenin pathway was detected.Results:The cell proliferation rate was lower than that of the control group and the apoptosis rate was higher than that of the control group at each time point in the low,medium and high dose groups,with statistically significant differences(P<0.05).The cell proliferation rate of berberine in the high dose group was lower than that of the low and medium dose groups,and the apoptosis rate was higher than that of the low and medium dose groups of berberine,with statistically significant differences(P<0.05).The number of cell invading cells and migrating cells in the low,medium and high dose groups of safranin were lower than those in the control group,and the difference was statistically significant(P<0.05).The number of cell invading cells and migrating cells in the high dose of berberine group were lower than those in the low and medium dose of berberine group,and the difference was statistically significant(P<0.05).The expression of Bax,Caspase3 and Caspase9 in cells of the low,medium and high dose groups of berberine was higher than that of the control group,and the expression of Bcl-2 was lower than that of the control group,and the difference was statistically significant(P<0.05).The expression of Bax,Caspase3 and Caspase9 in the high dose group of berberine was higher than that in the low and medium dose groups of berberine.The expression of Bcl-2 in the high dose group was lower than that in the low and medium dose groups,and the difference was statistically significant(P<0.05).The expression of Wnt,β-catenin,and VEGF protein in the cells of the low,middle and high dose groups was lower than that of the control group,and the difference was statistically significant(P<0.05).The cellular Wnt,β-catenin,and VEGF protein expressions in the high dose group of were lower than those in the low and medium dose groups,and the differences were statistically significant(P<0.05).Conclusion:Berberine intervention can promote apoptosis in prostate cancer cells,and its mechanism may be related to the regulation of Wnt/β-catenin signaling pathway.
