摘要
目的观察复方铁皮石斛对自发性高血压大鼠(SHR)的降压作用和对心肌组织中TRPV1 mRNA表达的影响。方法将24只SHR大鼠随机分为复方铁皮石斛低剂量组0.7 g/(kg·d)(低剂量组)、复方铁皮石斛中剂量组2.1 g/(kg·d)(中剂量组)、复方铁皮石斛高剂量组3.5 g/(kg·d)(高剂量组)、模型组,每组6只,5只WKY大鼠为正常组。各组大鼠经相应药物灌胃8周后分别测定给药后3 h、24 h血压,实时荧光定量PCR检测心脏TRPV1的基因表达。结果治疗第8周给药3 h、24 h后,与模型组比较,各治疗组血压均显著降低(P<0.01,P<0.05);与低剂量组比较,中剂量组及高剂量组降压幅度显著增加(P<0.01,P<0.05);与正常组比较,模型组TRPV1 mRNA表达显著升高(P<0.05);复方三个剂量组合计(复方高+中+低组)进行统计,TRPV1 mRNA表达较模型组显著下降(P<0.05)。结论复方铁皮石斛对SHR大鼠有显著降压疗效,其作用机制可能与降低SHR大鼠的TRPV1转录水平有关。
Objective To observe the antihypertensive effect of Compound Dendrobium Officinale(CDO)on spontaneously hypertensive rats(SHR)and the effect of expression of TRPV1 mRNA in heart tissue.Methods SHR rats were randomly divided into low-dose group of CDO0.7 g/(kg·d)(CDOL group),medium-dose group of CDO 2.1 g/(kg·d)(CDOM group),high-dose group of CDO 3.5 g/(kg·d)(CDOH group)and model group,with 6 rats in each group,and five Wistar-Kyoto(WKY)rats were taken as normal group.Systolic blood pressure(SBP)was measured in each group of rats at 3 h and 24 h after administration.The relative mRNA expressions of Transient Receptor Potential Vanilloid Type 1(TRPV1)were estimated by Realtime fluorescence quantitative PCR of heart.Results At 3 h and 24 h after administration,SBP in all treatment groups were significantly lower than that in the model group(P<0.01,P<0.05).The decreasing amplitude of SBP in CDOM group and CDOH group were significantly higher than that in CDOL group.The mRNA expression of model group was significantly higher than that in normal group(P<0.05).Compared with model group,the expression of TRPV1 mRNA was significantly decreased in the three groups with different doses of CDO.Conclusion Compound Dendrobium Officinale has significant antihypertensive effects on SHR and its mechanism may be related to the down-regulation of transcription of TRPV1.
