摘要
重组酶介导扩增(RAA)是重组酶等温扩增技术中极具代表性和前沿性的检测技术,弥补了现有核酸体外扩增技术需要热循环的局限性,使得核酸体外扩增更加简便快捷。RAA技术利用重组酶、脱氧核糖核酸(DNA)聚合酶和单链结合蛋白替代了传统的聚合酶链式反应(PCR)热循环解链过程,可在常温下完成扩增过程,同时兼具特异性强、灵敏度高、操作简单、耗时短等特点。该文在简述RAA技术的基础上,重点阐述了其在寄生虫、病毒、细菌、动物源性成分、植物病原等生物安全检测领域的应用,并对其未来发展前景予以展望,以期为拓展该技术的应用领域提供理论参考。
Recombinase-aid amplification(RAA) is a representative and advanced detection technique for recombinase isothermal amplification, which makes up for the limitation of thermal circulation of the nucleic acid amplification technology in vitro, and makes the nucleic acid amplification in vitro simpler and faster. RAA technology uses recombinase, DNA polymerase and single-stranded DNA binding to replace the traditional polymerase chain reaction(PCR) thermal cycling unchain process which can complete the amplification process at room temperature, and has the characteristics of strong specificity, high sensitivity, simple operation and short time consuming. Based on a brief introduction of RAA technology, the application in the biosafety detection of parasites, viruses, bacteria, animal-derived components and plant pathogens were briefly introduced, and its future development was prospected, in order to provide theoretical reference for expanding the application fields of the technology.
