干酪乳酪杆菌Zhang VBNC态在不同复苏基质中的代谢物组学研究

摘要 利用超高效液相色谱-四级杆飞行时间质谱技术(UPLC-Q-TOF MS)检测分析了干酪乳酪杆菌Zhang(Lacticaseibacillus casei Zhang,L.casei Zhang)VBNC态在液体MRS培养基、脱脂乳二种复苏基质中的代谢物差异。结果显示:在正离子和负离子模式下分别检测到2735种和1717种代谢物质。采用Metaboanalyst 5.0及SIMCA 14.1软件对代谢物进行分析,结合P<0.05、差异倍数>2和VIP≥1相结合的条件,共筛选出差异代谢物22种,归属于氨基酸、糖类、维生素、脂类和嘌呤类5大类,其中,甘氨酸、L-谷氨酰胺、低聚木糖和烟酰胺等18种代谢物在液体MRS培养基中的相对含量显著高于脱脂乳,其余4种(S-核糖基-L-高半胱氨酸、乳糖、核黄素和(R)-3-羟基丁酸酯)代谢物在脱脂乳中相对高些,并从成分角度研究分析了这些代谢物在L.casei Zhang VBNC态复苏中的可能作用,为初步探讨L.casei Zhang VBNC态的复苏机理提供了参考。 Ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF MS)was used to detect and analyze Lacticaseibacillus casei Zhang(L.casei Zhang)metabolome differences of VBNC state in liquid MRS medium and skimmed milk resuscitation substrates.Results showed that 2735 and 1717 metabolites were detected in positive and negative ion modes,respectively.The metabolites were analysed by Metaboanalyst 5.0 and SIMCA 14.1 software.Depending on the screening principles of P<0.05,Fold Change>2,and VIP≥1,a total of 22 different metabolites were screened.It belongs to 5 categories of amino acids,sugars,vitamins,lipids and purines.The relative contents of 18 metabolites(glycine,L-glutamine,xylooligosaccharide and nicotinamide,etc.)in liquid MRS medium were significantly higher than those in skim milk,and the other 4 metabolites(S-ribosyl-L-homocysteine,lactose,riboflavin and(R)-3-hydroxybutyrate)in skim milk were higher than in liquid MRS.The role of these metabolites in the recovery of L.casei Zhang VBNC state was analyzed from the perspective of composition,which provides a reference for the mechanism of recovery of L.casei Zhang VBNC state.
