摘要
试验旨在研究刺五加多糖纳米乳与鸡法氏囊灭活苗配合对雏鸡生长性能和免疫的影响,筛选刺五加多糖纳米乳作为佐剂对鸡法氏囊灭活苗的最适免疫增强剂量。选用1日龄AA肉鸡140只,随机分为7组,每组4个重复,每个重复5只鸡,ASPS-NE(L)组鸡注射32 g/L刺五加多糖纳米乳0.2 mL,ASPS-NE(M)组鸡注射64 g/L刺五加多糖纳米乳0.4 mL,ASPS-NE(H)组鸡注射128 g/L刺五加多糖纳米乳0.6 mL,单独法氏囊疫苗组每只鸡注射0.3 mL法氏囊疫苗,空白对照组每只鸡注射0.3 m L生理盐水,阳性对照组鸡注射10 g/L黄芪精0.4 m L,空白纳米乳组鸡注射0.4 mL纳米乳,所有组均在第14 d进行首次免疫,21、28 d进行二免、三免。试验期35 d。结果显示:与纯疫苗组、空白对照组、空白纳米组相比,ASPS-NE(M)组、阳性对照组雏鸡的平均日增重均极显著提高(P<0.01),料重比极显著降低(P<0.01)。将刺五加多糖纳米乳作为佐剂与鸡法氏囊灭活疫苗配合使用能够促进鸡法氏囊灭活疫苗抗体产生,增加雏鸡的免疫器官指数以及白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)的表达量,以中剂量刺五加多糖纳米乳效果最佳。研究表明,刺五加多糖纳米乳作为鸡法氏囊灭活苗的免疫佐剂具有良好的免疫促进作用,浓度为64 g/L时具有较好的免疫增强效果。
The experiment was to study the growth performance and immune-enhancing effects of Acanthopanax senticosus polysaccharide nanoemulsion combined with chicken bursal inactivated vaccine on chicks,to screen the optimal immune-enhancing dose of Acanthopanax senticosus polysaccharide nanoemulsion as adjuvant on chicken bursal inactivated vaccine.A total of 140 one-day-old AA broilers were randomly divided into seven groups,with four replicates in each group and five broilers in each replicate.The broilers in ASPS-NE(L)group were injected with 0.2 m L of Acanthopanax senticosus polysaccharide nanoemulsion(32 g/L),the broilers in ASPS-NE(M)group were injected with 0.4 mL of Acanthopanax senticosus polysaccharide nanoemulsion(64 g/L),the broilers in ASPS-NE(H)group were injected with 0.6 m L of Acanthopanax senticosus polysaccharide nanoemulsion(128 g/L),the bursal vaccine alone group was injected with 0.3 m L bursal vaccine,the blank group was injected with 0.3 mL normal saline,the positive control group was injected with 10 g/L Astragalus extract 0.4 mL,and the chickens in blank nanomilk group were injected with0.4 mL nanomilk.All groups were immunized for the first time on 14 d,followed by two immunizations and three immunizations on 21 and 28 d.The experiment lasted for 35 d.The results showed that compared with pure vaccine group,blank control group and blank nano group,the average daily gain of chicks in ASPS-NE(M)group and positive group was extremely increased(P<0.01),and feed to gain ratio was extremely decreased(P<0.01).Acanthopanax senticosus polysaccharide nanoemulsion as adjuvant combined with chicken bursal inactivated vaccine could promote the production of antibodies to chicken bursal inactivated vaccine,increase the immune organ index and the expression level of IL-2,IL-4 and IFN-γof chicks.Medium dose of Acanthopanax senticosus polysaccharide nanoemulsion had the best effect.The experiment indicates that the nano-emulsion of Acanthopanax senticosus polysaccharide has a good immune-promoting effect as an immune adjuvant of chicken bursa of Bursa inactivated vaccine,and the concentration of 64 g/L has a good immuneenhancing effect.
