摘要
目的:观察不同剂量的细辛水煎液对炎症与疼痛模型小鼠的影响,探讨不同剂量细辛水煎液的抗炎镇痛作用与肾毒性作用。方法:昆明小鼠180只,分3批分别进行醋酸扭体、耳廓肿胀和热板法实验,每批60只小鼠按体质量依据随机数字表随机分为对照组、阳性药组(给予地塞米松1.5 mg/kg)以及细辛水煎液剂量1、2、3、4组(给药剂量分别为24、12、6、3 g/kg),每组10只,雌雄各半。对照组小鼠按20 mL/kg体积灌胃蒸馏水,其余各组小鼠给予对应药物,每日1次,连续7 d。结果:与对照组相比,阳性药组小鼠耳廓肿胀度显著降低、扭体次数显著减少、痛阈值显著升高(P<0.01或P<0.05);细辛水煎液剂量1、2组小鼠耳廓肿胀度显著降低、扭体次数显著减少、痛阈值显著升高(P<0.01或P<0.05),细辛水煎液剂量3、4组小鼠耳廓肿胀度显著降低、痛阈值显著升高(P<0.01);细辛水煎液剂量1、2组小鼠肾组织中肾小管与集合管轻度扩张,皮髓质交界处少量的肾小管上皮细胞水肿,细胞肿胀,胞质疏松淡染。结论:24、12、6、3 g/kg剂量的细辛水煎液均具有显著的抗炎、镇痛作用,但24、12 g/kg剂量的细辛水煎液有轻微的肾毒性,而6、3 g/kg剂量的细辛水煎液对肾脏影响较小。
Objective:To observe the effects of Asarum decoction with different doses on inflammation and pain model in mice,thus to discuss different doses of Asarum decoction on anti-inflammation,analgesia and nephrotoxicity.Methods:A total of 180 mice were randomly divided into three patches for acetic acid writhing experiment,xylene-induced auricular swelling experiment and hot plate experiment.60 mice in each experiment were randomly divided into the control group,the positive drug group(Dexamethasone,1.5 mg/kg)and the Asarum decoction groups of 1,2,3 and 4(24 g/kg,12 g/kg,6 g/kg,3 g/kg),with 10 mice in each group.The control group was given 20 mL/kg distilled water by gavage.The other groups were given corresponding medications through intragastric administration at the dose of 20 mL/kg,once a day for consecutive 7 days.The degree of auricle swelling,the number of body twisting and the pain threshold of mice were measured,respectively.After that,the mice were sacrificed,and the renal tissues were collected for HE staining to observe the pathological changes.Results:Compared with those in the control group,the degree of auricle swelling and the number of twisting significantly decreased,and the pain threshold significantly increased in the positive drug group(P<0.01,P<0.05);the degree of auricle swelling and the number of twisting significantly decreased and the pain threshold significantly increased in the Asarum decoction groups of 1 and 2(P<0.01,P<0.05);the degree of auricle swelling significantly decreased and the pain threshold significantly increased in the Asarum decoction groups of 3 and 4(P<0.01).The renal tubules and collecting tubes were slightly dilated in the Asarum decoction groups of 1 and 2,and a small amount of renal tubular epithelial cells were swelling around the junction of renal cortex and renal medulla,and cytoplasm was loose and light stained.Conclusion:Asarum decoction has significant anti-inflammatory and analgesic effects at the doses of 24 g/kg,12 g/kg,6 g/kg and 3 g/kg,but the decoction has slight nephrotoxicity at the doses of 24 g/kg and 12 g/kg,and the decoction has little effect on kidneys at the doses of 6 g/kg and 3 g/kg.
