摘要
目的三阴性乳腺癌(triple negative breast cancer,TNBC)缺少有效治疗靶点,长链非编码RNA(long non⁃coding RNA,lncRNA)在TNBC发生发展中起着重要作用,本实验旨在探究lncRNA SPINT1⁃AS1对TNBC细胞增殖与凋亡的影响,寻找治疗TNBC的潜在靶标。方法qRT⁃PCR检测SPINT1⁃AS1在正常乳腺上皮细胞系以及TNBC细胞系的表达量;荧光原位杂交法检测SPINT1⁃AS1细胞内定位;构建SPINT1⁃AS1稳定过表达和低表达的TNBC细胞株,克隆形成实验、CCK⁃8实验、流式细胞术检测SPINT1⁃AS1对TNBC细胞增殖和凋亡的影响。结果SPINT1⁃AS1主要表达于胞质,在TNBC细胞中表达量显著低于正常乳腺上皮细胞,差异有统计学意义(P<0.05);SPINT1⁃AS1能显著下调细胞增殖能力(P<0.05),促进细胞凋亡(P<0.05)。结论SPINT1⁃AS1在TNBC细胞中低表达并且主要表达于细胞质;它能抑制TNBC细胞增殖并促进其凋亡。这提示SPINT1⁃AS1是TNBC潜在的治疗靶标。
Objective Triple negative breast cancer(TNBC)is an aggressive cancer without effective therapeutic targets.Long non⁃coding RNA(lncRNA)has been shown to play an important role in the development of TNBC.This study is intended to investigate the effects of lncRNA SPINT1⁃AS1 on the proliferation and apoptosis of TNBC cells and find a potential therapeutic target for TNBC.Methods The relative expression of SPINT1⁃AS1 in normal breast epithelial cell lines and TNBC cell lines was detected by real⁃time quantitative PCR(qRT⁃PCR).The cell localization of SPINT1⁃AS1 was detected by RNA fluorescence in situ hybridization.Overexpressed and down⁃regulated SPINT1⁃AS1 of TNBC cell lines were constructed respectively,and the proliferation and apoptosis of TNBC were explored by clone formation assay,CCK⁃8 assay and flow cytometry.Results SPINT1⁃AS1 was expressed mainly in the cytoplasm.The expression of SPINT1⁃AS1 in the TNBC cells was significantly lower than that in normal breast epithelial cells(P<0.05).SPINT1⁃AS1 downregulated the proliferation of TNBC cells in a significant way(P<0.05),and promoted their apoptosis(P<0.05).Conclusion SPINT1⁃AS1 is expressed at a lower level in the cytoplasm of TNBC cells.It can suppress the proliferation of TNBC cells and induce their apopto⁃sis,which indicates that it can be a potential therapeutic target for treating TNBC.
