摘要
为了能更好地利用畲药海金沙多糖,文章在单因素的基础上,利用响应面法优化微波传统和微波辐射预处理两种提取工艺,结合邻苯三酚自氧化法对提取物进行抗氧化研究。结果显示:微波传统提取的最佳工艺条件为微波功率280 W、微波时间57 s、液料比24(m L/g),在此条件下海金沙多糖提取率为7.756%,其提取物对O清除率为61.017%;微波辐射预处理提取的最佳工艺为微波功率350 W、微波时间71 s、液料比28(m L/g)、汽化剂用量5 m L,在此条件下海金沙多糖提取率为13.509%,其提取物对O清除率为63.089%。实验表明:微波辐射预处理可以有效地提高海金沙多糖的提取率,同时,海金沙多糖抗氧化活性有必要深入挖掘。
To make better use of the polysaccharide of Lygodium japonicum (Thunb.) Sw.,response surface methodology was used on the basis of single factor to optimize two extraction processes including microwave traditional and microwave radiation pretreatment.Pyrogallol autoxidation method was used to study the antioxidant activity of the extract.The results showed that the optimum conditions of microwave traditional extraction were microwave power 280 W,microwave time 57 s,liquid-material ratio 24 (m L/g);the extraction rate o polysaccharide with these conditions was 7.756%,and the Oscavenging rate of the extract was 61.017%;the optimum extraction technology of microwave radiation pretreatment were microwave power 350 W,microwave time71s,liquid-material ratio 28 (m L/g),vaporizer dosage 5 mL,and the extraction rate of polysaccharide with these conditions was 13.509%,and the Oscavenging rate of the extract was 63.089%.This experiment shows that microwave radiation pretreatment can effectively improve the extraction rate of total polysaccharides of L japonicum.At the same time,polysaccharides from L.japonicum on antioxidant activity needs to be further explored.
