基于RNA-seq筛选影响猪肉品质的关键候选基因 被引量:4

摘要 本研究旨在探究影响从江香猪和大白猪肉质差异的关键候选基因。实验以90日龄的从江香猪和大白猪为对象,利用转录组(RNA-seq)技术分析2个猪种背最长肌中差异基因表达谱,对差异表达基因(DEGs)进行GO和KEGG富集分析。qRT-PCR进行验证和筛选出肉质调控的关键功能基因。结果表明:生物信息学分析筛选出2个猪种DEGs 318个,其中上调基因有184个,下调基因有134个;GO分析发现,富集最显著的前30个GO条目,主要参与生物过程和细胞组分功能;KEGG分析发现,富集最显著的4条通路均与肉质相关,包括一磷酸腺苷活化蛋白激酶信号通路(AMPK Signaling Pathway)、转录因子信号通路(FoxO Signaling Pathway)、丙酮酸代谢(Pyruvate Metabolism)和脂肪酸代谢途径(Fatty Acid Metabolim)。通过筛选与验证,得到影响2个猪种肉品质的关键候选基因,其中同属硒蛋白家族的硒蛋白W(SelW)和硒结合蛋白1(SBP1)在2个品种猪中具有不同的表达模式,SelW在大白猪中高表达,而SBP1在从江香猪中高表达。本研究基于RNA-seq技术揭示了大白猪和香猪背最长肌中基因表达谱的差异,筛选肉质形成相关的信号通路和关键功能基因,为猪肉品质的分子调控机制奠定了基础。 This experiment was conducted to explore the key functional genes affecting longissimus dorsi muscle of Congjiang Xiang pigs and Large White pigs.In this study,90-day-old Congjiang Xiang pigs and Large White pigs were used to analyze the differential gene expression profile in longissimus dorsi muscle of the two breeds by RNA-seq technology,and the differential gene expression(DEGs)was analyzed by GO and KEGG enrichment analysis,and verified by qRT-PCR to screen out the key functional genes regulating meat quality.The results showed that through bioinformatics analysis,318 DEGs were screened out from Xiang pigs and LW pigs,including 184 up-regulated genes and 134 down-regulated genes.And GO analysis showed that the first 30 most significant GO items were mainly involved in biological processes and cell component functions,while KEGG analysis showed that the four most significant pathways of enrichment were all related to meat quality,including AMPK signaling pathway,FoxO signaling pathway,Pyruvate metabolism pathway and Fatty acid metabolism.Through screening and verification,the key candidate genes affecting the meat quality of two pig breeds were obtained.Selenoprotein W(SelW)and Selenium-binding Protein 1(SBP1),which belong to the same selenoprotein family,had different expression patterns in the two pig breeds.SelW was highly expressed in Large White pigs,while SBP1 was highly expressed in Congjiang Xiang pigs.To sum up,based on RNA-seq technology,this study revealed the differences of gene expression profiles in longissimus dorsi between large white pigs and Xiang pigs,and screened the signal pathways and key functional genes related to meat quality formation,which laid a foundation for the molecular regulation mechanism of pork quality.
